Trial 1 Study on the adsorptive ability of EGM to DON in vitroThe Esterified Glucomannan (EGM) which was synthesized in our laboratory was used to adsorb DON in aqueous phase, the solution of 500μg/ L DON was prepared, EGM was not added in the first and second group, 0.05%, 0.075%, 0.1%and 0.15% EGM was respectively added in the third group, the forth group, the fifth group and the sixth group at pH2.5, 0.05%,0.075%,0.1%and 0.15% EGM was respectively added in the seventh group, the eighth group, the ninth group and the tenth group at pH6.5, placed in the condition of 37±0.5℃for 12 hours, centrifugalizing at 5000 rounds per minute for 10 minutes. Detect the contents of DON, which could study the ability of EGM to adsorb DON in vitro when pH was 2.5 and 6.5. The result showed that the synthetical EGM had the satis ability of adsorption to DON, and the best adsorption rate was 35.54% when pH was 6.5 and the concentration of EGM achieved 0.1%.Trial 2 Effect of the adsorption of EGM to DON on cultured spleen cells in vitroThe spleen cells were cultured in vitro were divided into five groups for seventy-two hours to study the adsorption of EGM to DON , RPMI-1640 was chosen as basal culture medium, nothing was added in the first group; 500μg/L DON was added in the second group; 0.05%, 0.10% and 0.15% EGM was respectively added in the third group, the forth group and the fifth group. The result showed that the survival rate of spleen cells, the activity of SOD of the first group, the forth group and the fifth group were significantly higher than the second group (P<0.01), higher than the third group; the transformation rate of lymphocyte of the forth and the fifth group were higher than the second group and the third group (P<0.05), the content of MDA in the second group and the third group was higher than the first group, the forth group and the fifth group (P<0.05), and the difference of all indices among the forth group and the fifth group were not significant(P>0.05). It concludes that EGM can effectively adsorb DON and relieve the toxic effect of DON to cultured spleen cells in vitro, and the best additive dose of EGM in diet is 0.10%.Trial 3 The protective effect of EGM on mice fed with the diets contaminated by DONTo studied the effect of EGM added in the feed contaminated by DON toxin on the toxication prevention of DON toxin, one hundred and fifty healthy male Kunming mice were randomly divided into five groups for thirty days. The first group was fed with basal diet. The second group was fed with basal diet added 2mg/kg DON toxin. The third group, the forth group and the fifth group were fed basal diet added with 2 mg/kg DON toxin and 0.05%, 0.10%, 0.15% EGM respectively. The results showed that, the average daily weight gain of mice, the spleen and the thymus indices of the first group, the forth group and the fifth group were significantly higher than the second group (P<0.05); the number of erythrocyte and leucocyte and the content of hemoglobin and the transformation efficiency of lymphocyte of the first, the forth and the fifth group were significantly higher than the second group(P<0.01); Liver indices of the second group were significantly higher than the first, the forth and the fifth group(P<0.01). the difference the biochemical parameters among the first, the forth and the fifth group were not significant(P>0.05). the content of MDA, the activity of AST and ALT of the second and the third group were higher than the first, the forth and the fifth group (P<0.05); the activity of SOD, LDH and GSH-Px of the first, the forth and the fifth group were significantly higher than the second group(P<0.01), higher than the third group (P<0.05); the difference the biochemical parameters of sereum among the first, the forth and the fifth group were not significant(P>0.05). It concludes that EGM can effectively adsorb DON toxin, preventing the mice from the injury of DON toxin, and the the proper additive dose of EGM in diet is 0.10%.
