Konjac is a special cash crop containing konjac glucomannan in large quantities.The shorter growth period and lower yield of konjac cause the longer dormancy stage.The study on the low temperature of 4 ? after harvest Amorphophallus muelleri corm was treated in this experiment,and use some physiological methods to measure each philology indexes.Such as weight method,double wave length method,methyl catechol method,NBT Illumination method et al.Get the following results:1.In the process phase of low temperature treatment,the change of free water content was parabolic.During low temperature storage,the ratio of free water to total water was increasing,and the ratio to 40 d reached the highest.The total water content in the tissue fluctuated up and down,and the water content was the highest at 30 d,then it fell down,and then maintains remained a relatively stable state.The increase of the ratio of free water / total water content can be used as one of the signs of the dormancy released of the corm of A.muelleri.2.The process of corm dormancy release was accompanied by vigorous activities of carbohydrate metabolism.Cold storage can promote amylase activity and promote starch hydrolysis.During storage period,starch content and soluble sugar content were increasing.The content of soluble sugar in different parts of the tissue changed in different types,and the content of soluble sugar in the whole was higher than that before treatment.Among them,the most obvious changed in bud tissue,and the parenchyma continued to rise in 0-30 d,the highest state at 30 d;the content changes of peripheral tissue without excessive volatility,had been in a relatively stable state,but also at 30 d was the highest.The starch content decreased,the apical bud tissues decreased significantly,the lowest in 50 d;there was a fluctuation in the parenchyma during 0-20 d,20-50 d continued to decline,50-60 d rebound;smaller changes in peripheral tissue.The change trend of soluble protein and soluble sugar was almost consistent.The change trend of starch and soluble sugar was opposite,the starch decreased but the soluble sugar increased,which had a positive effect on the germination of tissue.3.The activities of various metabolic enzymes in different tissues were different.Overall,cold treatment can improve the antioxidant enzyme activities,effectively remove the damage of reactive oxygen species to plant cells,effectively promote the release of corm dormancy,and provide an appropriate material basis for subsequent bulb germination and seedling growth.The changes of the three enzymes of SOD,POD and PPO were low during cold storage,and the highest point of their activity appeared at different periods.Bud tissue during 0-40 d,SOD activity in the parenchyma tissue and peripheral tissue are increased to the maximum at 40 d.The activity of POD fluctuates greatly,and the activity was maximum at 20 d,and then decreased.The activity of PPO reached the highest at 30 d.Through the above results deepens our understanding of dormancy physiology of A.muelleri corms during cold storage,and provides important physiological parameters of dormancy released.It can provide reference to seek for the better and faster way of Amorphophallus muelleri corm dormancy release.
