Livestock and poultry diarrhea is one of the most important diseases that endanger the breeding industry.Chinese medicine is the treasure of our country and plays a key role in the prevention and cure of human and animal diseases.The natural,low residual and low drug resistance of Chinese veterinary drugs are unmatched by chemical drugs,and are the inevitable trend of green healthy breeding.Probiotics are active microorganisms that are beneficial to the host,and are commonly used to treat gastrointestinal related diseases.Some data show that some Chinese medicine and probiotics combined treatment of intestinal diseases is better than single use of traditional Chinese medicine or probiotics alone.In this study,the combination of Chinese herbal medicine purslane,cordatehouttuynia and probiotics was used to prevent diarrhea in animals.It is a useful exploration for the combination of traditional Chinese medicine and probiotics.In this study,we used mice as experimental animal,purslane andcordate houttuynia as tested drugs,56 strains of probiotics as tested strains and test the following.?1?Probiotics screening.Probiotics was screened by artificial gastrointestinal fluid tolerance test,inhibition of enterotoxigenic Escherichia coli test and effects of purslane and cordate houttuynia on the growth and reproduction of probiotics.?2?Animal experiment.Different doses?1,2.5 and 5 g/kg?of purslane and cordate houttuynia were added to the basal diet of mice to prepare the medicated feed firstly and the shape of medicated feed was the same as that of normal mice feed.240 male Kunming mice were randomly divided into 15 groups,namely:different dose group of purslane?1,2.5,5 g/kg?,different doses of purslane?1,2.5,5 g/kg?and probiotics group,different dose group of cordate houttuynia?1,2.5,5 g/kg?,different doses of cordate houttuynia?1,2.5,5 g/kg?and probiotics group,probiotics group,model group and blank group.The concentration of probiotics in each group was 1×108CFU/mL,and theintragastric dose was 0.2 mL/10g weight once a day.The medicine was giving all the time.In addition to the blank group,10d was administered to the rest of the mice and the model was constructed.?4?Construct a mouse diarrhea model.Dry ginger water extract?0.5 g/mL?was administered at a dose of 0.28mL/10g body weight once a day;after 10 d,intraperitoneal injection of at 0.2 mL/10g weight,concentration of 7.5×107 CFU/mL ETEC once a day,continuous 3 d.At the end of the test and at the end of the test,the rate of diarrhea in mice was recorded and the biochemical indexes of blood were detected,and the intestinal histopathology was observed.The inflammatory cytokines including TNF-?,IL-1?,IL-6,IL-10,TGF-?1,IFN-?were detected by fluorescence quantitative PCR in intestinal tissue.Result:?1?Probiotics screening 7 strains of probiotics were screened out from the 58strains of probiotics,which were able to tolerate the environment of artificial gastrointestinal fluid,which were composed of 3 strains of yeast,2 strains of lactic acid bacteria,and 2 strains of Bacillus subtilis.Through Synergistic effect of probiotics by purslane and cordate Houttuynia and Antagonistic effect of probiotics on pathogenic Escherichia coli.,3 strains were further screened out from the 7 strains of probiotics,which were composed of 1 strain of yeast,1 strain of lactic acid bacteria,and 1 strain of Bacillus subtilis.?2?Histopathological changes The pathological model showed that the small intestine of the model group was broken,the epithelial cells were swollen,some of them were necrotic,the inflammatory cells were increased and the intestinal glands were deformed obviously.Compared with the model group,the mice in the treatment group were restored,and the intestinal villi in the high dose group were basically intact and the inflammatory cells were decreased.?3?Diarrhea rate In the model group,the diarrhea rate was 87.5%and was higher than that of other groups in the first day.The diarrhea rate of cordate houttuynia in each medication group was higher than that of the same dose of purslane.Compared with the purslane group,the diarrhea rate of mice in high dose purslane combined with probiotics group was the lowest?12.5%??4?Electrolyte detection Compared with the blank group,the content of Cl-in serum of model group increased significantly,and the contents of Ca2+decreased significantly.Compared with the model group,the mice in each group were able to restore the physiological function of mice to some extent,and the effect of purslane probiotics group was better than that of other groups.?5?Detection of inflammatory related factors Compared with the blank group,the expression of TNF-?,IL-1?,IL-10,IL-6,TGF-?1,IFN-?were significantly increased in model group?P<0.05?,indicating that ETEC infection caused by intestinal injury in mice with ginger,causes the related factor of each inflammatory bowel tissue was increased.Compared with the model group,the expression of purslane probiotics in high dose group IL-1?,IL-6,IL-10,TNF-?,IFN-?,TGF-?1 was significantly decreased?P<0.05?,show that the expression of anti-inflammatory cytokines up-regulated,down regulated expression of pro-inflammatory cytokines to resist ETEC with ginger on the body of the infection and prevention of diarrhea.Conclusion:?1?ETEC combined with dried ginger can induce heat toxin syndrome diarrhea,cause electrolyte imbalance,intestinal injury in mice.?2?The combined application of purslane or cordate houttuyniawith probiotics can reduce the heat toxin syndrome diarrhea rate,and the combination effect of purslane-probiotics is better than that of cordatehouttuynia-probiotics.?3?The combination effect of purslane and probiotics can restore electrolyte balance and reduce intestinal inflammation by changing IL-1?,IL-6,TNF-?,IFN-?,TGF-?1,IL-10 inflammatory factors reactionin heat toxin syndrome mice.
