The purpose of this paper is to study the protective effects of royal jelly against carbon tetrachloride-induced liver injury in rats by proteomics.The main contents and results of this research are summarized as follows:1.The study was designed to establish the model of carbon tetrachloride-induced liver injury in rats:Three different CCl4-induced hepatic injury models were studied by observing growth conditions and determining the level of serum AST and ALT of rats.Model 1:rats were received 50%CC14 corn oil(2 mL · kg-1)in the first day.Model 2:rats were received 50%CCl4corn oil(1 mL · kg-1)every other day,for 6 times.Model 3:rats were received15%CC14 corn oil(2 mL · kg-1)in the first,forth,seventh and tenth day,for 4 times.Compared with normal group,the level of AST and ALT of the model group in model 1 after receiving CCl424h and 48h significantly increased,which not significantly increased after 72 h and one week.The result showed probable self-healing.Compared with normal group,the serum AST and ALT of the model group in model 2 and model 3 significantly increased.There were rats died during experiment in model 2.Because of the exorbitant infected rate,the therapeutic status could not be reflected.Therefore we selected the model 3 as experimental model.2.The study was designed to evaluate the protective effects of royal jelly on carbon tetrachloride-induced liver injury in rats.Based on the preliminary study,a further study about the protective actions of royal jelly in carbon tetrachloride-inducedliver injury in rats was investigated from four sides(the liver index,the ALT,AST and ALP in rat serum,the methane dicarboxylic aldehyde(MDA)in liver homogenate,and liver tissue slice).Compared with normal group,the level of liver index,ALT,ALP,MDA in model group significantly increased(P<0.01),while the level of AST significantly increased(P<0.05).Compared with model group,the level of liver index,ALT,AST and MDA in royal jelly(0.4 g·kg-1)group significantly reduced(P<0.01,P<0.01 P<0.05,P<0.05),and the level of ALP in royal jelly(0.4 g ·kg-1)group was between nomarl group and model group,and the level of liver index?ALT?AST?MDA?ALP in royal jelly(0.8 g·kg-1 and 1.6 g·kg-1)group significantly reduced(P<0.01,P<0.01,P<0.01,P<0.01 and P<0.05).The results of liver histopathology showed that:compared with normal control group,we found edema lesions and severe inflammatory cell infiltration in model group,and these phenomena were improved in each group of royal jelly,which further confirmed the above results.3.Proteomics analysis of the effects of royal jelly on carbon tetrachloride-induced liver injury in rats was studied.The protein in rat liver of different experiment group(nomarl group,model group and royal jelly group)was studied by optimizing the two-dimensional gel electrophoresis experiment condition(selection of IPG,loading quantity of sample),and analyzing the picture.The result showed that:compared with normal and royal jelly group,there were up-regulated 42 proteins and downed-regulated 25 proteins.The proteins with significantly different expression were identified by MALDI-TOF/TOF-MS,and 32 proteins were successfully determined.The proteins that up-regulated in model group than nomal group and royal jelly group were:phospholipase C I,Osteogenic growth peptide,D-dopachrome decarboxylase,Serum albumin,Cytokeratin-18,Peroxiredoxin-2,40kDa ribosomal protein,Heat shock 70 kDa protein 9,Aldoketomutase,3-mercaptopyruvate sulfurtransferase,alpha-tropomyosin 5b,60 kDa heat shock protein,Heat shock protein 90 kDa beta member 1,and the proteins that downed-regulated in model group than normal group and royal jelly group were:Phosphatidylethanolamine-binding protein 1,Regucalcin,probable 10-formyltetrahy-drofolate dehydrogenase ALDH1L2,Actin,succinate dehydrogenase(ubiquinone)flavoprotein subuni,cytokeratin 8 polypeptide.The results showed that most of determined proteins were associated with metabolism,and some proteins(such as cytokeratin 18,heat shock proteins)that can be used as bio-marker of the protective effect of royal jelly on carbon tetrachloride-induced liver injury in rats.The results revealed the pathways and mechanisms of the protective effect of royal jelly on liver damage in some degree.
