Objective:Investigating the effect of Phyliathus Complex in different periods of rehabilitation of carbon tetrachloride-induced liver injury in canines,to provide an effective method of treatment for pets clinical liver injury rehabilitation.Methods:12 one year old healthy adult canines(weighing 8?10kg)were randomly divided into three groups,namely:carbon tetrachloride model group,carbon tetrachloride and biphenyl dimethyl dicarboxylate(DDB)group,carbon tetrachloride and Phyliathus Complex group(referred to as:model group,the control group,TCM group),n=4.The first four weeks,all the canines were in accordance with 1.2mL/kg dose gavage 50%carbon tetrachloride solution rapeseed oil,once a week for a total of 4 times.Fifth to eighth week,model group provides only basic diet and water;the control group added DDB 0.2mg/kg on the daily basic diet;TCM group added aqueous extract of Phyliathus Complex with 1mL/kg dose on the daily basic diet.Before the test and trials,weekly B-scanning,and collected venous blood samples to conduct blood routine and blood biochemistry(AST,ALT,ALKP,TP,ALB,MDA,GSH,T-SOD),performed abdominal exploration and biopsy of liver tissue to HE staining,and observe the pathological changes,after the end of the trial.Results:1.Effect of Phyliathus Complex on carbon tetrachloride-induced liver injury in canines blood routine indicatorsThe number of white blood cells and red blood cells,hematocritduring,during the test canines in each group,always maintained in the normal range and no significant variation.The first week of the test values for each group canine platelets appear significantly increased,including TCM group and the model group was significantly higher among the model group(P<0.05)and the control group had no significant difference(P>0.05),the second to the fourth week of platelet values continue to decrease,the fourth week when surrounded by each group was not significantly different(P>0.05),the fifth week to the eighth week of each group increased the number of platelets but remained at a healthy adult canines of the reference range in which the medicine was significantly higher than the model group(P<0.05),the control group was significantly higher than that of TCM group(P<0.05).2.Effect of Phyliathus Complex of liver function on carbon tetrachloride-induced liver injury in caninesCompared with the previous test,the first four weeks,ALT,AST,ALKP significant increasing trend indicator;TP,ALB trend indicators appear to reduce the difference between changes in these indicators in each group of canines were not significant(P>0.05).Fifth to eighth week,ALT,AST,ALKP of TCM group and the control group indicators have a clear downward trend,in the control group and Chinese medicine group was significantly lower than the model group(P<0.05),and the control group was significantly lower than that of TCM group(P<0.05).TP,ALB trend indicators have significantly increased,there was no significant difference between the groups(P>0.05).3.Effect of Phyliathus Complex for antioxidant indicators on carbon tetrachloride-induced liver injury in caninesBefore the test,each group canine GSH and T-SOD activity were not significantly different(P>0.05),the control group was significantly lower than the activity of canine MDA model group and Chinese medicine group(P<0.05),model group and medicine between groups were not significant(P>0.05).The fourth week T-SOD indicators have significantly increased,GSH and MDA indicators have decreased,and there were no significant differences between these indicators canine in each group(P>0.05).The fifth to eight weeks,Chinese medicine group and the control group T-SOD and GSH indicators showed a clear downward trend,MDA indicators was significantly increased trend indicators.TCM group which canine GSH and T-SOD activity was significantly lower than the control group(P<0.05),the control group GSH and T-SOD activity was significantly lower than the model group(P<0.05),the active group was significantly higher canine MDA(P<0.05),MDA activity in the control group was significantly higher than the model group(P<0.05).4.Effect of Phyliathus Complex on carbon tetrachloride-induced liver injury in canines liver tissue structureThe first four weeks,ultrasound imaging show canine in each group had scattered patchy parenchymal echogenicity in the liver portal system expansion filling,siltation and even gallbladder cholecystitis images.Fifth to eighth week canine model group ultrasound imaging show the liver parenchyma patchy,scattered echogenic liver edge blur irregular echogenic image sediment samples occur within the thin edge blunt gallbladder,occupies nearly the entire gallbladder.ultrasound imaging parenchymal canine in the control group scattered echogenic rounded edges of the gallbladder shape increases,wall thinning,gallbladder unclean sediment samples hyperechoic image appears inside,accounted for almost two-thirds of the gallbladder.TCM group ultrasound scattered over image parenchymal echogenicity,sharp edges,portal system clear,gallbladder cleanse,normal size.After the test,abdominal exploration discovery model group showed scattered canine liver necrosis,rounded edges,hard and brittle texture,pathology observed in liver cell degeneration and necrosis leading to liver plates arranged chaos,degeneration of liver cells enlarged liver sinusoidal formation of oppression sinusoid so narrow,some areas appear congestion necrosis,a large number of connective tissue proliferation phenomenon occurs,these changes especially obvious central vein area.TCM group and the control group,the above performance significantly reduced liver damage,pathology was not observed in the central vein thickening,liver cells showed only slight variability,no significant periportal connective tissue proliferation phenomenon.Conclusion:Phyliathus Complex significantly reducing enzyme action,as well as a significant antioxidant effect,can effectively repair the liver cell degeneration after injury,reduce the incidence of liver connective tissue proliferation,prevention of fibrosis,is clinically effective Hepatoprotective veterinary medicine.
