Acute liver injury is an acute liver lesion caused by physical,chemical,biological and other pathogenic factors.In veterinary clinic,there is currently no specific drug treatment for acute liver injury or chronic liver failure,and active supportive treatment and symptomatic treatment,early diagnosis and early treatment are still emphasized,with poor overall prognosis.In recent years,stem cell transplantation has shown great potential and advantages in human medicine.However,in veterinary clinic,there is not much research.This article aims to study the therapeutic effect of adipose-derived mesenchymal stem cells?ADMSCs?on acute liver injury in dogs caused by carbon tetrachloride?CCl4?and explore new drugs for the treatment of liver injury diseases in dogs.In this experiment,a safe and reliable model of CCl4acute liver injury in dogs was first established.Twenty dogs of similar age and weight?about 8 kg?were selected and randomly divided into four groups,with 5 dogs in each group.A was the control group,B,C and D were the test groups.In the test groups B,C and D,50%CCl4peanut oil solution?CCl4:peanut oil=1:1?was administered intraperitoneally at 0.5 ml/kg,1.0 ml/kg and 1.5 ml/kg body weight,respectively.In group A,no drugs were used.The clinical symptoms and biochemical indexes of each group were monitored for 7 consecutive days,and the pathological changes of liver tissues on the 3rd and 7th day of each model were monitored.Secondly,the therapeutic effect of ADMSCs on CCl4-induced acute liver injury in dogs was studied and compared with that of common symptomatic treatment.Fifteen dogs with acute liver injury successfully modeled with 1 ml/kg dose of 50%CCl4?CCl4:peanut oil=1:1?were randomly divided into three groups,with 5 dogs in each group.group e was the control group,group f and g were the experimental group,group e was untreated,group f was ADMSCs group of dogs transplanted via vein,group g was the common symptomatic treatment group.the clinical symptoms and biochemical indexes of each group were monitored for 10 days continuously,and the pathological changes of liver tissues on the 5th and 10th days of treatment in each group were monitored,and the data were analyzed.The test results show that:1.There were no abnormal changes in clinical symptoms,biochemical indexes and liver histopathology in group A dogs during modeling.The clinical symptoms,liver biochemical indexes and liver histopathological changes of dogs in group B gradually returned to normal after modeling,which may be that the dose of CCl4is small and the degree of damage to the liver is lower than the liver's self-repairing ability.In group C,the clinical symptoms changed,liver biochemical indexes were abnormal,liver histopathology also changed,and no dog died.The clinical symptoms,liver biochemical indexes and liver histopathology of test dogs in group D were abnormal during the modeling period,resulting in death of test dogs.The analysis may be due to excessive modeling dose,resulting in death of test dogs.That is,1ml/kg intraperitoneal injection of 50%CCl4peanut oil solution can establish a stable and reliable acute liver injury model in dogs.2.During treatment,clinical symptoms,biochemical indexes and histopathology of test dogs in group E continued to deteriorate,and 4 dogs died.During the treatment of test dogs in group F,the clinical symptoms of 5 test dogs were relieved without death,serum ALT and AST decreased significantly on the 2nd day respectively,and liver histopathology improved.During the treatment of test dogs in group G,the clinical symptoms of 2 test dogs were relieved without death,serum ALT and AST decreased significantly on the 2nd to 3rd d respectively,and liver histopathology did not improve significantly.That is,ADMSCs transplanted via vein has therapeutic effect on acute liver injury caused by CCl4in dogs and is better than common symptomatic treatment.To sum up,1.A stable and reliable model of acute liver injury in dogs can be established by intraperitoneal injection of 50%CCl4peanut oil solution at 1ml/kg.2.ADMSCs transplanted via vein has therapeutic effect on acute liver injury caused by CCl4in dogs,and is better than common symptomatic treatment.
