Abstract/Summary:
Currently,cancer is still one of the major killers of harming human health,most of the cancer drugs not only target cancer cells,but also harm the normal cells,so seeking out a new low toxicity,targeted drugs will become an active area of Bioinorganic Medicinal Chemistry.Platinum as anti-cancer drugs have been resistanced by some tumors,and them also cause the patient vomiting,liver,kidney and other organ damage.So the development of anticancer drugs of transition metal complexes targeting non-covalent DNA binding becomes a focus.Although the organic metal complex for a long time been considered to be unstable in physiological conditions,the organometallic complexes still provide tremendous opportunities for the design of new anticancer drugs.Studies have demonstrated that the aryl group ruthenium complexes,C,N-ring metal of Ru,Rh,Ir,Pt complexes showed a certain anti-cancer activity.In addition,Pd complexes which has similar structural and thermodynamic properties with Pt complexes also shows the potential anticancer properties.Therefore,this paper designed synthesis of a series of new ring metal palladium complexes containing rimantadine derivative Schiff base and studied the interaction between the series of complexes and bovine serum albumin,which lay the foundation for the research on anti-cancer activity of the series of compounds.Meanwhile,in order to carry out a comparative study,we only synthesized Pd complexes of rimantadine derivative Schiff bases ligands,and its structure was completely characterized.The major contents of this paper are as follows:We first synthesized[Pd{(4-R)C6H4CH=NC6H3-2,6-i-Pr2}(μ-Cl)]2(R= H,1;R =OMe,2)which has been reported in our previous work.Then we use complex 1 and 2 with schiff bases derived from rimantadine to synthesize ring palladium complexes[Pd(C,N-K2-C6H4CH=NC6H3-2,6-i-Pr2){N,O-k2-C12H19N=CH-(2-O-C6H4)}](3),[Pd(C,N-k2-C6H4CH=NC6H3-2,6-i-Pr2){N,O-k2-C12H19N=CH-(3-MeO-2-O-C6H4)}](4),[Pd(C,N-k2-C6H4CH=NC6H3-2,6-i-Pr2){N,O-k2-C12H19N=CH-(4-MeO-2-0-C6H4)}](5),[Pd(C,N-K2-C6H4CH=NC6H3-2,6-i-Pr2){N,O-k2-C12H19N=CH-(5-Cl-2-O-C6H4)}](6),[Pd{C,N-k2-(4-MeO)C6H3CH=NC6H3-2,6-i-Pr2}{N,C)-k2-C12H19N=CH-(2-O-C6H4)}](7),[Pd{C,N-k2-(4-MeO)C6H3CH=NC6H3-2,6-i-Pr2} {N,O-k2-C12H19N=CH-((3-MeO-2-O-C6H4)}](8),[Pd{C,N-k2-(4-MeO)C6H3CH=NC6H3-2,6-i-Pr2} {N,O-k2-C12H19-N=CH-(4-MeO-2-O-C6H4)}](9),[Pd{C,N-k2-(4-MeO)C6H3CH=NC6H3-2,6-i-Pr2}{N,O-k2-C12H19N=CH-((5-Cl-2-O-C6H4)}](10).All these complexes were fully characterized by FT-IR,1H NMR spectroscopy and elemental analysis.The molecular structure of 3,5 and 8 were determined by the X-ray crystallography.Bovine serum albumin(BSA)as a target protein,the interaction of compounds 3-10 and BSA was studied by using fluorescence spectroscopy,synchronous fluorescence spectra spectroscopy.Synthesis and characterization of palladium complexes of rimantadine and its derivative schiff base:we use rimantadine salicylaldehyde and its derivatives Schiff bases reacted with Na2PdCl4,these palladium complexes which were coordinated by Pd-N and Pd-O.The complexes are Pd[k2-N,O-C12H19N=CH-(2-O-C6H4)]2(11),Pd[k2-N,O-C12H19N=CH-(3-MeO-2-O-C6H4)]2(12),Pd[k2-N,O-C12H19N=CH-(4-MeO-2-O-C6H4)]2(13),Pd[k2-N,O-C12H19N=CH-(5-Cl-2-O-C6H4)]2(14).Then we get all single crystal of all these compounds.All these complexes were fully characterized by FT-IR,1H NMR spectroscopy and elemental analysis.The molecular structures were determined by the X-ray crystallography.
