Objectives:The purpose of this study was to obtain basic health data of Han residents in the cold plateau region of Yunnan Province,and to understand the distribution of PPARy2 gene Prol2Ala(rs 1801282)and C161-T(rs3856806)polymorphism population in the Han plateau regions of the Yunnan plateau.To explore the polymorphism of PPARy2 gene rs1801282 and rs3856806 loci in Han residents in plateau cold regions,and to provide a new idea for the early prevention,diagnosis and treatment of obesity.Methods:The data of this subject comes from the National Key R&D Program of China topic "Study on the Cohort of Natural Populations in High Altitude Areas in Southwest China".The cohort adopts the sampling method of cluster sampling to determine the research site and research objects.Demographic information,life behavior habits,dietary intake,nutritional status,disease history and other basic conditions,and basic health data and blood biochemical indicators of Han residents in cold plateau areas were obtained from questionnaire,medical examinations and laboratory tests.Among the 10,544 people in the baseline survey,they were grouped according to BMI level:BMI?28kg/m2 was the obese group(475 cases),BMI was 26.9-27.9 kg/m2 was super-recombination(284 cases),BMI was 18.5-23.9 kg/m2 is the normal group(759 cases).Then the obese group and the obese group matched the normal group(normal group 1),super-recombination and super-recombination matched normal group(normal group 2)according to age(±2),gender,and township 1:1 matching,a total of 1518 cases.Genomic DNA was extracted using a small kit of AxyPreP blood genomic DNA,and SNaPshot method was used to genotype the PPAR?2 gene rs1801282 and rs3856806.All data analysis was carried out with statistical package of social sciences(SPSS)23.0 software to analyze the association between obesity and PPARy2 gene rs1801282 and rs3856806 polymorphisms.Statistical analyses were conducted by t test,chi-square test,rank sum test,conditional logistic regression analysis,and P<0.05 was considered to be statistically.Results:1.A total of 10,544 Han people in Yongsheng County,Lijiang City,Yunnan Province were actually surveyed on the site.Among them,526 were obese(4.99%),and 2523 were overweight(23.93%).A total of 1,518 people were included in the study,of which 475(31.29%)were obese,284(18.71%)were overweight,and 759(50%)were normal controls.2.Genotypes,and combined genotypes of the PPAR?2 gene rs1801282 and rs3856806 loci in the plateau Han obesity group and the obesity group matched the normal group(normal group 1),super recombination and super recombination matched normal group(normal group 2).There was no statistically significant difference(P>0.05).3.The obesity group and the obesity group matched the normal group(normal group 1).The mutation rate of rs1801282 locus was 10.74%and 11.16%respectively,and the gene frequencies were(CC genotype:89.26%vs 88,84%,CG genotype:10.74%vs 11.16%;C allele:94.63%vs 94.42%,G allele 5.37%vs 5.58%);rs3856806 mutation rates were 42.94%and 44.21%respectively,and the gene frequencies were CC genotype:57.05%vs 55.79%,CT genotype:37.68%vs 38.53%,TT genotype:5.26%vs 5.68%;C allele:75.89%vs 75.05%,T allele 24.11%vs 24.95%);the combined genotype frequencies of rs1801282 and rs3856806 are(CC+CC genotype:54.74%vs 53.90%,CC+CT genotype:31.37%vs 30.53%,CC+TT genotype:3.16%vs 4.42%,CG+CC genotype:2.31%vs 1.89%,CG+CT genotype:6.32%vs 8.00%,CG+TT genotype:2.10%vs 1.26%).4.Super recombination and super recombination matched normal group(normal group 2)The mutation rate of rs1801282 locus was 7.04%and 9.86%respectively,the gene frequency was(CC genotype:92.96%vs 90.14%,CG genotype:6.34%vs 9.51%,GG genotype:0.70%vs 0.35%;C allele:96.13%vs 94.89%,G allele 3.87%vs 5.11%);rs3856806 mutation rates were 44.01%and 40.14%,the gene frequencies were(CC genotype:55.99%vs 59.86%,CT genotype:38.38%vs 36.62%,TT genotype:5.63%vs 3.52%;C allele:75.18%vs 78.17%,T allele 24.82%vs 21.83%);rs1801282 and rs3856806 combined genotype frequencies were(CC+CC genotype:54.58%vs 58.45%,CC+CT genotype:33.10%vs 29.23%,CC+TT genotype:5.28%vs 2.46%,CG+ CC genotype:1.41%vs 1.41%,CG+CT genotype:4.93%vs 7.04%,CG+TT genotype:0.00%vs 1.06%,GG+CT genotype:0.35%vs 0.35%,GG+TT genotype:0.35%vs 0.00%).5.Super-recombinant matched normal group(normal group 2)CC genotype,C allele carriers mostly do not drink,gender,age,smoking at Prol2Ala(rs1801282)site genotype,allele differences are all no statistical significance(P>0.05).6.Super-recombinant matched normal group(normal group 2)CG genotype HDL is higher than CC genotype and GG genotype,other clinical indicators have no statistically significant difference between CC genotype,CG genotype and GG genotype(P>0.05);the BMI of CC genotype and CT genotype in obese group was higher than that of TT genotype,but there was no significant difference in other clinical indicators between CC genotype,CT genotype and TT genotype(P>0.05).7.Conditional logistic regression results of obesity and overweight of Han residents in plateau indicate that high levels of DBP,GLU and LDL are the risk factors for obesity;smokers,high levels of SBP and TC are the risk factors for overweight;high HDL is the protective factors for overweight and obesity.Conclusions:1.The polymorphisms of rs1801282 and rs3856806 of PPAR?2 gene may not be related to the risk of obesity of Han people in the cold region of Yunnan Plateau.2.The normal group(normal group 2),super-recombinant and super-recom binant matched normal group(normal group 2)of the Han nationality obese gr oup and the obese group in the cold region of Yunnan Plateau,the polymorphi sms of the PPARy2 gene rs1801282 and rs3856806 are all The C allele is the dominant gene and distributed among the Han population in the plateau cold region.The homozygotes are mainly CC genotypes,the mutant genotypes are mainly heterozygous mutations CG and CT genotypes,and the homozygous mu tation GG genotypes are in Only 3 were found in this study population,and t here were few homozygous mutant TT genotypes,among which TT genotype may have a beneficial effect on obesity.3.High levels of DBP,GLU and LDL may be the risk factors for obesity;smoking,high levels of SBP and TC may be the risk factors for overweight;high HDL may be a protective factor for obesity and overweight.
