Waterlily is a perennial herbaceous flower of Nymphaeaceae.According to the ecological characteristics,it can be divided into cold-resistant waterlily and tropical waterlily.As a precious flower and an important waterscape greening material in aquatic flowers,waterlily is favored and preferred by people because of its brilliant and changeable color,sacred and lofty moral,improvement of water eutrophication,strong adsorption ability to heavy metals and other harmful substances in water and silt.In addition,the flower of waterlily can be made into scented tea,and the pedicel and petiole of waterlily can also be edible,which is full of edible and ornamental value as well as waterscape and landscape value.In order to make full use of the abundant hydrothermal resources in Guangxi and promote the basic research of aquatic flowers,especially waterlily,the adaptability evaluation and genetic diversity analysis of more than 40 waterlily germplasm resources collected from home and abroad were carried out.The results are as follows:(1)Apparent traits and adaptability of 49 waterlily germplasm resources collected from home and abroad were determined and evaluated by means of statistics and principal component analysis.Evaluation systems and standards for apparent traits were established.Thirty-nine varieties(species)which were suitable for planting in Guangxi were selected preliminary from all samples,accounting for 79.6%of the total number of introduced varieties.This shows that most of the introduced varieties(species)grow well in Guangxi,among which 16 varieties(species)including Nymphaea 'Eidorado:Nymphaea micrantha,and Nymphaea rubra are the most worthy of further popularization and application in Guangxi.(2)10 primers with high polymorphism and good repeatability were screened out from 100 ISSR primers and a genetic diversity analysis system of waterlily based on ISSR markers was constructed then.A total of 281 bands were amplified from 46 waterlily germplasm resources,with an average of 28.1 bands per primer,281 polymorphic bands and 100%polymorphic ratio.The average Shannon information index(?)was 0.4197,and the average Nei's gene diversity index(H)was 0.2657,and the average Observed allele number(Na)was 2.000,and the average Effective allele number(Ne)was 1.4139,which showed abundant genetic diversity.The genetic similarity coefficient was between 0.51 and 0.98.Based on the genetic similarity coefficient,a clustering tree was constructed to reveal the genetic relationship of 46 Waterlily germplasm resources.When the similarity coefficient was 0.68,all the tested materials could be clustered into six groups,which supports the classification of the Nymphaea L.into five subgenuses according to their geographical origins.(3)Based on ISSR molecular marker technology,DNA fingerprints of 24 primary species of waterlily collected from home and abroad were constructed.According to the differences of DNA fingerprints,different primary species of waterlily could be identified easily and quickly.At the same time,it can also provide molecular technical supports for the classification,identification and protection of new germplasms and varieties of waterlily.
