Zizania latifolia is a genus of the genus Oryzae,a subfamily of the genus Oysteraceae.It is an aquatic herb.According to reports in the literature,there are four species of genus,namely Zizania latifolia,Zizania texana,Zizania aquatica and Zizania palustris.As the most primitive species of the genus Amaranthus,the Zizania latifolia has strong resistance to deep water,drought resistance and disease resistance,including resistance to bacterial blight,sheath blight and rice blast,and has water purification and heavy metals for polluted water bodies.Enriched ecological restoration capacity.As one of the closely related genus of rice,Zizania latifolia has many excellent genes and has high research value.It plays an important role in the improvement of rice germplasm and is an ideal resource for enriching the genetic resources of crop breeding.In recent years,genetic research on Zizania latifolia is gradually deepening.SSR markers have the advantages of rich information content and high polymorphism,and are important molecular markers for studying genetic diversity.Studies have shown that rice SSR markers can be used for genetic analysis of Zizania latifolia,but the marker is still in a blank stage in the study of genetic diversity across the country.Therefore,9 pairs of rice SSR primers were screened,and 381 cockroach materials of Zizania latifolia from 25 a populations of Zizania latifolia in China were tested by PCR amplification to analyze their genetic diversity.The main findings are as follows:(1)9 pairs of rice SSR primers were screened,SSR-PCR reaction system was optimized,381 Zizania latifolia sputum materials were amplified by PCR,and PCR amplification products were detected by 6% polyacrylamide gel electrophoresis.A total of 119 clear and stable samples were amplified.The band,wherein the polymorphic strip carries 113,and an average of 11.44 polymorphic sites are amplified by each pair of labeled primers.The average polymorphic locus ratio of 9 pairs of SSR primers in rice can be as high as 94.77%,and the ratio of polymorphic loci in 4 pairs of rice SSR primers is as high as 100%.(2)The percentage of polymorphic loci(PPL)in 25 populations of Zizania latifolia in China was 94.50% at the species level,Nei's gene diversity index(H)was0.2398,and Shannon's diversity information index(I)was 0.3772,indicating that Zizania latifolia in China is rich in genetic diversity;genetic variation among Chinese Zizania latifolia cockroaches(50.79% of total variation)is more obvious than genetic variation within population(49.21% of total variation),genetic diversity among populations of Zizania latifolia in China The(Ht)was 0.2310,and the genetic diversity(Hs)in the population was 0.1122.The genetic diversity among the populations of Zizania latifolia in China was more abundant than that in the population.The genetic differentiation coefficient(Gst)among the populations of Zizania latifolia in China was0.5144.The gene flow(Nm)between groups was 0.4720(Nm<1),and genetic drift played an important role in the genetic differentiation of Zizania latifolia populations.(3)The genetic similarity coefficient(Gs)and genetic distance(Gd)of 25 populations of Zizania latifolia in China ranged from 0.7693 to 0.9725 and 0.0279 to0.2623,respectively.The correlation between genetic distance and geographical distance of 25 populations of Zizania latifolia in China was r=0.8331,p=0.001**,which was significantly correlated.At the threshold of 0.88,the genetic structure of 25 populations of Zizania latifolia in China The clustering can be divided into 5 categories,and the clustering results are related to the geographical distribution,and there is a certain correlation.In the evolutionary relationship,25 populations of Zizania latifolia in China are divided into two branches by the common ancestors,the first The branches first differentiated from Guilin(GL)in the central and southern regions,and the second branch first differentiated from the Dongting Lake(DTH)in the central and southern regions.It is speculated that the Zizania latifolia in the central and southern regions are relatively primitive,and Zizania latifolia in China locusts began to evolve from the central and southern regions.
