In this paper, ive representative geographical population of Lilium pumilum DC including 153 strains as materials were studied. For instance, the Baota District of Yan'an City, Ansai County, Fuxian, Huangling County and Yanchuan County were selected. We used the morphology(seven Shape indexes like bulb diameter, plant height, stem diameter, leaf length, leaf width, leaves number, and flowers number were used) method and inter-simple sequence repeat(ISSR) molecular marker technology to discuss the Lilium pumlium DC genetic diversity of germplasm resources in Yan'an. In order to investigate the genetic relationship between the wild germplasm resources in Yanan area of Lilium pumilum DC, the cluster analysis of Lilium pumlium DC between these five representative geographical populations of Yan' an were performed. Further, the correlation analysis between climatic factors(maximum and minimum annual air temperature, annual mean temperature, above sea level, frost-free season, annual sunshine, mean annual precipitation) and genetic diversity was performed for clarifying the effects of geographical, climatic factors on genetic diversity of germplasm resources of Yan'an wild Lilium pumlium DC. The results were shown as follows:1 Through the phenotypic studies of five Lilium pumilum DC wild populations, we found that wild Lilium pumilum DC is widely distributed in Yan' an, five samples are suitable for the growth of Lilium pumilum DC habitat, and the morphology characteristics including bulb diameter, stem height, stem base color, leaf length and leaf width are different from the Flora of China. These results indicated that germplasm resources of wild Yan' an Lilium pumilum DC has its specificity. Morphological characters showed that the difference among populations is greater than that among populations. The main differences come from the plant height and leaf length.2 The results of correlation analysis between morphological shape and geographical factors are as follows: Firstly, bulb diameter and frost free period have significantly negative correlation and bulb diameter have significant correlation with annual sunshine hours. Secondly, height of plant height and altitude have significantly negative correlated height of plant height have extremely significant correlation with annual sunshine hours or average annual rainfall. Thirdly, the stem diameter is significantly correlated with the average annual precipitation. Fourthly, the number of leaves is significantly correlated with the annual sunshine hours, and it is significantly correlated with the average annual precipitation. Lastly, the number of flowers was significantly negatively correlated with the annual maximum temperature, and was significantly correlated with the annual sunshine hours. Other correlations are not significant.3 Seven shape indexes(bulb diameter, plant height, stem diameter, leaf length, leaf width, leaves number, and flowers number) which were selected from many indexes of each population represented each population. And cluster analysis was done and results showed that five populations could be divided into 2 branches, Ansai County and Yanchuan County in the first branch was aggregated at first, and was aggregated with Baota District populations. The Fuxian and Huangling County population was aggregated in the second branch.4 Five primers with rich polymorphism were selected from 8 primers. And 5 wild populations were analysised by ISSR-PCR. 54 sites were detected, and the polymorphic rate was 77.78%. The population level genetic diversity index(The highest He's is Ansai County: 0.2941; the lowest He's is Yanchuan County: 0.1245); The polymorphic loci(PPL)at the population level(The highest is Huangling County: 71.24%; The lowest is Fuxian County: 42.58%). Baota District, Ansai County, Fu County, Huangling County and Yanchuan County has a certain level of genetic differentiation among populations.The difference among populations is greater than that among populations.5 The genetic distance(D) which counted by ISSR marker and the genetic identity(I) was inputted in NTSYS-pc 2.1 software, and UPGMA cluster analysis was performed in molecular level. The results showed that the genetic distance Ansai county population and in Baota District group had nearest genetic distance. Fuxian County and HuanglingCounty came second, the genetic distance of Yanchang county from other county was farthest, finally cluster with other population.6 Results of correlation analysis between ISSR marker genetic diversity and climate,geographical factors are as follows:There is no significant correlation between ISSR genetic diversity and climate, geographical factors. The correlation between different Lilium pumilum DC populations genetic diversity index and altitude, annual maximum temperature, minimum temperature, annual mean temperature, frost free period, the annual sunshine hours, and the average annual precipitation is not significant.
