Bletilla striata is a perennial herbaceous plant of Orchidaceae Bletilla genus.The bulbs can be used as medicine.Hemostasis,detumescence and sarcogenic convergence are regarded as main medicinal value.With the rapid development of Chinese medicine industry in recent years,people continuously develop the medicinal value of B.striata in most areas of China,which results in wild Bletilla resources have been on the verge of exhaustion.Rhizosphere bacteria microorganism can not only affect the yield and quality of medicinal plants,but also change the effective components of plants.Therefore,the study of the relationship between medicinal plants and bacteria microbes is beneficial to the realization of the target of high quality and high yield of medicinal plants.The thing that the seeds were sowed in different mediums was studied in this paper,and then buds and roots were inducted.Soil bacterial community composition and abundance of three samples from Jingde,Jinzhai and Wufeng counties were determined by high-throughput sequencing technology.They will contribute to illustrate the influence factors on the growth environment and the difference of bacterial microbial community structures in different areas.The infection rate of AMF in root of Bletilla striata from Jinzhai was studied by the acid fuchsin staining method.Wet sieving method was used to isolate spore.The spores of isolated AMF were identified by morphology method to discuss AMF diversity in B.striata soil.This paper provides theoretical bases for the cultivation and growth of Bletilla seedlings.The main results were as followings:1.In this experiment,seeds as materials were in the seeding medium of different hormone concentration and different culture to study the effect of different hormone concentration culture medium on seeds germination,bud induction and rooting induction.The experimental results showed that 1/2MS+NAA 1.0 mg/L was beneficial to seeds germination;MS+6-BA 1.0 mg/L+NAA 0.1 mg/L was beneficial to buds induction;1/2MS+NAA 0.2 mg/L+6-BA 0.1 mg/L was beneficial to roots growth.2.The soil bacteria in three regions were studied by high-throughput sequencing technology.124783 effective sequences were obtained from three soil samples.2906 OTUs were gained at the similarity level of 97%.The bacteria OTUs were mainly assigned into 28 known phyla,56 known classes and 295 known genera.There were 5 dominant bacteria(relative abundance > 5%)in the Jingde area,Proteobacteria,Bacteroidetes,Acidobacteria,Planctomycetes and Chloroflexi respectively;there were 6 dominant bacteria in the Jinzhai area,respectively Proteobacteria,Acidobacteria,Bacteroidetes,Actinobacteria,Planctomycetes and Chloroflexi;there were 6 dominant bacteria in the Wufeng area,respectively Proteobacteria,Cyanobacteria,Bacteroidetes,Acidobacteria,Actinobacteria and Chloroflexi.Proteobacteria,Bacteroidetes,Acidobacteria and Chloroflexi were the dominant bacteria in the three regions.3.AMF in rhizosphere soil of Bletilla had clear hyphae and vesicles,and the infection rate was above 90%.4 groups and 25 species of fungi were isolated from rhizosphere soil of B.striata by optical microscopy and electron microscopy.In which 9 species were in Acaulospora,14 in Glomus,1 in Gigaspora and 1 in Scutellaspora.Glomus and Acaulospora were the dominant genera.
