Glaciers as a representative of the extreme environment occupied about 10% of the area of the earth.There are some microbe which have unique microbial genetics and mechanism for adapt to environmental change.It aslo reflected the bacteria quantity and bacterial community structure changes.the reserach of glacial microorganisms not only is a way to develop the extreme environment resources,it is also a important way to explore the relationship between microorganisms and environmental changes.By the methods of pure culture and high-throughput sequencing,bacteria diversity was investigated in glacial of west kunlun and Xiong Caigangri area.We have been achieved main result as follows :1.Total number of culturable bacteria is about 1.75×104CFU/m L-1~2.21×104CFU/m L-1in Xiongcaigangri region glacier snow samples,1.46×104~3.73×104CFU/m L-1in water samples,Pure culture results showed that glacier bacteria community was composed of Proteobacteria ?Bacteroidetes ? Actinobacteria ? Deinococcus-Thermus at phylum level,It was composed of Pseudomonas ? Janthinobacterium ? Flavobacterium ? Devosia ? Duganella ? Deinococcus ?Cryobacterium?Rugamonas?Arthrobacter?Mycetocola?Massilia?Salinibacterium at genus level.2.High-throughput sequencing results showed that xiongcaigangri area glacial bacteria community was composed of Proteobacteria ? Bacteroidetes ? Firmicutes ? Acidobacteria ?Actinobacteria?Candidate_division_TM7?Chloroflexi?Cyanobacteria?Gemmatimonadetes ?Planctomycetes?Verrucomicrobia at phylum level.It was mainly composed of Acidovorax,Bacillus,Bacteroides,Brevundimonas,Herbaspirillum,Massilia,Pedobacter,Phenylobacterium,Polaromonas,Pseudorhodobacter,Variovorax at genus level.The Snow sample bacteria community diversity is greater than water sample.3.Bacterial diversity was similar between glacial snow and glacial Water,It is not similar with previous studies.4.The total number of culturable bacteria is about 2.7×102CFU/m L~2.27×104CFU/m L in Chongce glacier snow samples,7×104CFU/g in moraine sample,5.1×105CFU/g~1.59×106CFU/g in glacier foreland soil samples,Pure culture results showed that glacier bacteria community was composed of Actinobacteria?Proteobacteria?Bacteroidetes?Firmicutes at phylum level.It was composed of Geobacillus?Modestobacter ?Hymenobacter?Methylobacterium ?Mycobacterium ?Paracoccus?Bacillus?Janibacter?Lysinibacillus?Brevundimonas?Rhizobium?Aureimonas ?Kocuria?Nocardioides?Psychrobacter?Arthrobacter?Microbacterium?Mycetocola?Patulibacter?Rhodococcus?Massilia ?Aerococcus? Clavibacter?Xylophilus?Skermanella?Pseudomonas?Rugamonas?Paenisporosarcina?[Brevibacterium] frigoritolerans at genus level.5.High-throughput sequencing results showed that chongce glacial bacteria community was composed of Acidobacteria?Actinobacteria?Armatimonadetes?BRC1?Bacteroidetes?Chlorobi?Chloroflexi?Cyanobacteria?Elusimicrobia?FBP?Firmicutes?Fusobacteria?Gemmatimonadetes?GAL15?GN02?MVP-21?Nitrospirae?OD1?OP11?Planctomycetes?Proteobacteria?spirochaetes?TM7?Verrucomicrobia?WS3?[Thermi] at phylum level.It was mainly composed of Kaistobacter?Flavisolibacter?Spirosoma?Methylibium?Phormidium?Hymenobacter?Nitrospira?DA101?Gemmata?Rubellimicrobium?Thermomonas?Deinococcus?Rudanella?Lysobacter?Janthinobacterium?Leptolyngbya?Pseudanabaena?Rubricoccus?Rubrobacter?Rhodobacter?Sporocytophaga?Adhaeribacter?Roseococcus?Polaromonas?Leptothrix?Planctomyces?R18-435?Acinetobacter?Pseudonocardia?Ochrobactrum?Euzebya?Anoxybacillus?Corynebacterium?Propionibacterium?Rhodococcus?Bradyrhizobium?Methylobacterium?Sphingomonas?Pseudomonas at genus level.(relative abundance>1%)6.The microbial diversity and abundanceof chongce glacier in moraine is higher than soil,and soil is higher than snow,Microbial communities in the snow were significantly different from those in the moraine and glacier forland soil.Microbial communities in the snow differentsite is Quite different,but it is similar between different site soil samples.7.The results is not similar for two methods,it showed that the combination of culture-dependent method and high-throughput sequencing can obtain more comprehensive investigation of bacterial community.
