Study On The Motor Function Stimulation Parameters Of Rats With Sciatic Nerve Injury Treated By Three Methods And Three Points

[Objective]Through the orthogonal experiment under the guidance of comparative behavior,changing the chemical indexes of the gastrocnemius muscle tissue function,immune effect of massage,the parameters of the power,from time and frequency on motor function of rats with recovery of peripheral nerve injury.To explore the best combination of stimulus parameters for the recovery of motor function in rats with sciatic nerve injury.[Method]The sciatic nerve injury was taken as an example to study the peripheral nerve injury.The SD rats were used as experimental animals and the sciatic nerve injury model was used.7 days after the use of "massage Simulator" simulation method(three dial method,rubbing method,point massage method)in three points(yin men,Cheng shan,yang linq uan)on intervention.power(A),time(B),frequency(C)three factors,each factor has three different levels of stimulation,the power for 2N,4N,6N;time lmin,3min,5min;frequency 30 times/min,60 times/min,100 times.Using the orthogonal test design of three factors and three levels,the best stimulus parameters combination of massage intervention to promote the recovery of motor function in SNI rats was screened.According to the behavioristics,determination to evaluate the function recovery of rats tiltboard experiment and sciatic nerve function index1.From the morphological point of view,the gastrocnemius muscle was stained with HE to observe the morphology of muscle cells and wet weight of gastrocnemius muscle2.The expression of NGF in ventral horn of spinal cord was measured by immunohistochemical method,and the optimal stimulation parameters of NGF expression were explored.[Result]1.Inclined plate test:After 20 times of massage intervention,the massage group(nine groups)were analyzed by orthogonal experiment.The results showed that the relationship between the three factors was:time>strength>frequency.The best combination of stimulus parameters is power 4N,time 1min,frequency 60 times/min.The variance analysis was used in the massage group(nine groups),and the influence of three factors power(A),time(B)and frequency(C)on the result was not statistically significant(P>0.05).But from the F-measure size(orthogonal test in corresponding factors F-measure is larger,the impact of the larger,can be considered),the relationship between various factors:time>>power frequency,and better treatment effect in short time.This is consistent with the results obtained by the intuitive analysis2.Sciatic nerve function index test:After 20 times massage power 4N,time 5min,frequency 30/min.4.Expression of NGF in anterior horn of spinal cord:The tuina group and model group,there were statistical difference(P<0.05).After 20 times of tuina intervention,the tuina group,model group and normal group were statistical difference(P<0.05).After 20 times of tuina,tuina group(nine groups)was analyzed by orthogonal test,the results show that the primary and secondary relations of the three factors are power,time and frequency.The best combination of stimulus parameters is power 4N,time 1min,frequency 60/min.[Conclusion]1.Tuina can promote the recovery of motor function in SNI rats.According to the inclined plate test:and sciatic nerve function index,time and frequency factor the most significant impact on motor function recovery.The best stimulation parameters to promote motor function recovery,the inclined plate test is power 4N,time 1min,frequency 60 times/min..;the sciatic nerve function index is power 2N,time 1min,frequency 30 times/min.The results showed a certain range power medium,short time and uniform stimulation helps hind limbs motor function recovery,and light,soft,slow stimulation is more advantageous to digit movement function recovery.2.The tunai intervention can promote the SNI rat ventral horn of spinal cord NGF expression.The best stimulation parameters for power 4N,time 1min,frequency 60 times/min.
