Abstract/Summary:
[Objective]By detecting the changes of Cdh8proteinsexpressedin the dorsal horn of the spinal cord,TRPAlproteins,TRPM8proteins,Cdh8proteins,and PAR2 proteinsexpressed in the dorsal root gangliaand TRPA1proteinsexpressed in the skin epithelial cells in SNI model rat,we investigated the effection ofcold sensory hypersensitivityin peripheral nerve injurytreat with"three methodsand three points"tuinatherapy.[Method]Through establishingsciatic nerve injury modelrats and treat with "three methodsand three points"tuina intervention with them.And using immunohistochemical and western blot techniquesto qualitatively and quantitatively studyCdh8proteinsin rat spinal cord dorsal horn,TRPAl,TRPM8,Cdh8,PAR2in dorsal root ganglion and TRPA1in foot skin epithelial.cells.From the levelsofthe expression of the dorsal horn of spinal cord Cdh8 proteins,the expression ofTRPAlproteins,TRPM8proteins,Cdh8proteins,PAR2proteins of the dorsal root ganglions and the expression of Cdh8 proteinsin the epithelial cells of rat foot,and combining with the study of the cold sensory hypersensitivity behavior of the SNI rat using cold plate toexploreingthe effection ofthe SNI rat modelcold sensory hypersensitivity treat with "three methodsand three points" tuinatherapy.[Result]1.Cold plate experimentAt 1 day before modeling,there was no significant difference with the number of foot lifts in each group(P>0.05).With 0 day of tuina intervention,therewas no significantly.differencewith the number of foot lifts between the normal group and the sham group(P>0.05);Compared with the sham group,the number of foot lifts in the model group and the tuina intervention group increased significantly(P<0.01).With 20 day of tuina intervention,compared with the model group,the number of foot liftswithtuina intervention group decreased significantly(P<0.01).2.The expression ofCdh8 proteins in the dorsal horn of the spinal cordImmunohistochemistry assay and Western Blot testof Cdh8 proteinsexpressed in spinal dorsal horn neuronsresults showed that,with 0 day of tuina intervention,there was no significant difference in the expression of Cdh8 proteins between the normal spinal cord and the sham operated group(P>0.05).Compared with the sham group,the expression of Cdh8 proteinsin the dorsal horn of the model group increased significantly(P<0.05).With 20 day of tuina intervention,compared with the model group,Cdh8 proteinsexpressedin the spinal dorsal horn of the tuina intervention group rat reduced.3.The expression ofTRPAl,TRPM8,PAR2 and Cdh8 proteinsin the dorsal root ganglionImmunohistochemical assayand western blot test ofTRPAlproteins,TRPM8proteins,PAR2 proteinsand Cdh8 proteinsin the dorsal root ganglion showed that,with 0 day of tuina intervention,there was no significant difference in the expression of TRPA1 proteins,TRPM8proteins,PAR2proteins,Cdh8 proteins between the normal spinal cord and the sham operated group(P>0.05).Compared with the sham group,the expression of TRPA1 proteins,TRPM8proteins,PAR2proteins and Cdh8proteinsin dorsal root ganglion were increased in model group(P<0.05).With 20 day of tuina intervention,compared with the model group,TRPAlprotein,PAR2protein expressed in the spinal dorsal horn of the tuina intervention group rats reduced(P<0.05).However,the reducechanges of Cdh8proteinand TRPM8 protein were not obvious.4.The expression of TRPA1 protein in skin epithelial cellsImmunohistochemical assay of TRPAlin skin epithelial cells results showed thatthere was no significant difference in the expression of TRPAlproteins between the normal spinal cord and the sham operated groupafter 0 day of tuina intervention(P>0.05).Compared with the sham group,the expression of TRPA1 proteinsin skin epithelial were increased in model group(P<0.05).Compared with the model groupafter 20 days of tuina intervention,the expression of TRPA1 proteinsin the skin epithelial cellsof the tuina intervention group were decreased(P<0.05).[Conclusion]1."Three methods and three points" tuina therapy can significantly improve the cold sensory hypersensitivityreaction of SNI rats.2.The recovery mechanism of "three methods and three points" tuina therapy for cold sensory hypersensitivity in SNI rats isassiciatewith the regulation of the related cold receptor pathwayof TRPAlproteins.3.The recovery mechanism of "three methods and three points" tuinaintervention forcold sensory hypersensitivity in SNI rat wasworked though inhibiting the expression ofRPAlprotein and PAR2protein in dorsal root ganglion,and the expression of TRPA1 in skin epithelium.
