[Objective]To study the effect of the power,time and frequency of "three methods and three points" manipulation parameters on the sensory function of rats with sciatic nerve injury from the behavioral perspective.comparing the changes of the indexes under different parameters from the morphological and immunohistochemical perspective to explore the suitable parameter of tuina to promote the recovery of sensory function in rats with sciatic nerve injury.[Method]Taking sciatic nerve injury as an reference to study peripheral nerve injury,SD rats were used as experimental animals,sciatic nerve injury model was established.Using the "tuina simulator" to simulate the tuina technique,Using the orthogonal experiment design of three factors and three levels to explore the effects of different stimulation parameters on the sensory function of SNI rats.1.According to the.behavioristics,measure the thermal pain tolerance to evaluate the sensory function recovery of rats,From the functional point of view,the recovery rate of gastrocnemius wet weight was measured to evaluate the improvement of muscle atrophy.2.From the morphological point of view,the gastrocnemius muscle was stained with HE to observe the morphology of muscle cells and to measure the diameter of muscle cells.3.The expression of NGF in dorsal root ganglion and dorsal horn of spinal cord was measured by immunohistochemical method,and the optimal stimulation parameters of NGF expression were explored.[Result](1)Determination of resistance to thermal pain threshold7 days after modeling,The tuina group and model group thermal tolerance threshold determination of no significant difference(P>0.05).After 20 times of tuina intervention,The tuina group thermal pain threshold and determination of resistance results compared to the model group had significant difference(P<0.05).Sensory function recovery was close to sham operation group.After 20 times of tuina,tuina group(nine groups)was analyzed by orthogonal test,the results show that the primary and secondary relations of the three factors are power,frequency and time.The best combination of stimulus parameters is power 2N,time 5min,frequency 100/min.The tuina group(nine groups)was analyzed by variance analysis,the factors of A,B and C had significant difference(P<0.05),However,from a single factor,the influence of A factors was significant(P=0.0154),B and C factors were not significant(P>0.05).(2)Recovery rate of gastrocnemius wet weight7 days after modeling,There was no significant difference in the recovery rate of gastrocnemius wet weight between the tuina group and the model group(P>0.05),After 20 times of tuina intervention,Compared with the model group,the recovery rate of wet weight of gastrocnemius muscle in the tuina group was significantly different(P<0.05),The degree of muscle atrophy was close to that of sham operation group.After 20 times of tuina intervention,tuina group(nine groups)was analyzed by orthogonal test,the results show that the primary and secondary relations of the three factors are power,frequency and time.The best combination of stimulus parameters is power 4N,time 1min,frequency 30/min.The tuina group(nine groups)were analyzed by variance analysis,the factors A,B,C of the primary and secondary relations had no significant difference(P>0.05).(3)Muscle cell diameter measurement7 days after modeling,there were no significant difference between the tuina group and the model group(P>0.05),After 20 times of tuina intervention,there was a significant difference in muscle cell diameter between the tuina group and the model group(P<0.05),The diameter of muscle cells was close to that of sham operated group.After 20 times of tuina intervention,tuina group(nine groups)was analyzed by orthogonal test,the results show that the primary and secondary relations of the three factors are time frequency and power.The best combination of stimulus parameters is time 5min,frequency 100/min,power 4N.The tuina group(nine groups)were analyzed by variance analysis,the factors A,B,C of the primary and secondary relations had no significant difference(P>0.05).So there was no significant difference between the primary and secondary factors.(4)Expression of NGF in dorsal root ganglion7 days after modeling,there was no significant difference in the expression of NGF in the dorsal root ganglion between the tuina group and the model group(P>0.05).After 20 times of tuina intervention,the expression of NGF in the tuina group was higher than that in the model group(P<0.05).After 20 times of tuina intervention,tuina group(nine groups)was analyzed by orthogonal test,the results show that the primary and secondary relations of the three factors are frequency time and power.The best combination of stimulus parameters is time 3min,frequency 60/min,power 2N.The tuina group(nine groups)were analyzed by variance analysis,the factors A,B,C of the primary and secondary relations had no significant difference(P>0.05).So there was no significant difference between the primary and secondary factors.(5)Expression of NGF in dorsal horn of spinal cord7 days after modeling,the expression of NGF in dorsal horn of spinal cord in model group was increased(P<0.05).There was no significant difference between the tuina group and the model group(P>0.05).After 20 times of tuina intervention,the expression of NGF in the spinal dorsal horn of the tuina group was significantly different from that of the model group(P<0.05),Tuina intervention promoted the expression of NGF in dorsal horn of spinal cord.After 20 times of tuina intervention,tuina group(nine groups)was analyzed by orthogonal test,the results show that the primary and secondary relations of the three factors are power time and frequency.The best combination of stimulus parameters is power 4N time 1min,frequency 60/min.The tuina group(Group Nine)was analyzed by variance analysis which fonund that the factors A,B,C had significant difference(P<0.05).But from a single factor,A(power)factor,B(time)factors significantly(P<0.05),C(frequency)factors are not significant(P>0.05).[Conclusion](1)Manipulation of "three methods and three points" intervention can promote the recovery of sensory function in SNI rats.(2)Power is one of the key factors of tuina.mild strength,long time and high frequency stimulation is more conducive to the recovery of sensory function.(3)The mechanism of the effect of tuina on the recovery of sensory function in rats may be related to the expression of NGF in dorsal root ganglion and dorsal horn of spinal cord of SNI rats.
