Objective To explore the influence of buyang huanwu decoction (BYHWD) secretion of nerve growth factor (NGF) and mRNA expression on the peroneal nerve in rats after anterior tibial peroneal nerve injury, we hope that this study will contribute to the mechanism of Bu Yang Huan Wu decoction treatment of movement disorders caused by peripheral nerve damage and provide objective theory of clinical application and further research by traditional Chinese medicine.Methods1.Discussions on the theory treating movement disorders caused by nerve damage. Collect literature flaccidity zheng systematically and western medicine on peripheral nerve injury, in order to investigate the prevention and treatment of peripheral nerve injury in movement disorders and summarize recent relevant BYHWD,compatibility, single herbs and compound modern pharmacological research literature.2.Experimental study on the effect of Buyanghuanwu treatment of peroneal nerve injury resulting in movement disorders. 48 male Sprague-Dawley rats were subjected to common peroneal nerve crush model, then they were randomly divided into sham operation group, mecobalamin group (positive control),model group, BYHWD high-dose,medium-dose,low-dose groups.Each group has eight rats. The gavage lasted for 21 d. After that,toe width was measured, recording exhibition refers to the function; Then EMG testing were performed,measuring RNS decay rate; The wet weight ratio of tibial muscle was measured; Tibialis anterior muscle HE staining was measured, recording anterior tibial muscle fiber cross-sectional area; the use of transmission electron microscopy techniques to observe shape and thickness of myelin sheath; immun ohistochemical staining secretion in rat NGF protein and mRNA experession by PCR were measured. This observation Buyanghuanwu decoction administered to rats peroneal nerve regeneration and functional recovery.Results1.Movement disorder caused by the equivalent of the motherland medicine belongs to "flaccidity zheng" category. One of the main pathological mechanism of onset is battered, blood barrier. Disease resistance based virtual reality wrangling centered, blood stasis as the standard. The main treatment method is reinforcing qi and blood. As Buqihuoxue law on behalf of parties, BYHWD is the correct prescriptions.2.exhibition toe function test(TSF):After the first 2d ipsilateral toes curled and appeared limp.7d after BYHWD, the high-dose group and toe mecobalamin group gradually recovered and the model group’s recovery was very slow. The situation continued to improve after 14d.21d each group significantly extended toe gait returned to normal. But in addition to the sham group, the other groups affected different degrees of movement disorders. Compared with model group, BYHWD high-dose group, mecobalamin group are significant different(P<0.01 or P<0.05). And there is no significant difference between BYHWD groups and mecobalamin group (P> 0.05).3.wet weight ratio:Compared with sham group,the ipsilateral tibial muscle wet weight ratio has shown varying degrees decreasing.Compared with the model group,BYHWD high-dose, mecobalamin groups were increased obviously(P<0.05). While there exists no difference between BYHWD high dose and mecobalamin group (P> 0.05).4.RNS decay rate:In addition to the sham group,other groups’ RNS all decayed.Compared with model group,BYHWD high-dose group, mecobalamin group perform significant difference (P<0.01). Between high-dose and low-dose group,there exist important difference (P<0.01)5.tibialis anterior muscle cross-sectional area:Before the sham group tibial muscle cross-sectional shape than the rule, little connective tissue. Compared with sham group, model group muscle fiber structural disorder, the cell gap widened, connective tissue proliferation, degeneration of muscle fibers shrink significantly. Compared with sham group, BYHWD groups and group insurance indemnity can intercellular space widened, mild atrophy of muscle fiber degeneration, but more regular shape, structure is still clear, more uniform staining, connective tissue proliferation inconspicuous muscle fiber cross-sectional area Statistical analysis show compared with the sham group, the other five groups are significantly different (P<0.05). And there is no differemce between mecobalamin group and BYHWD high-dose group(P>0.05).6.The common peroneal nerve morphology electron microscope:In the sham group, axonal transection intrinsic axonal microtubules can be seen, neurofilament and shafting film. Myelinated part cross-section of the shape, the myelin lamellar structure neat, uniform, no discrete phenomenon.Change of BYHWD and mecobalamin group myelin sheath thickness is not obvious, only a small amount of myelin degeneration of nerve fiber layer of discrete plate. BYHWD, the low-dose group, further thinning the thickness of the myelin sheath, plies a discrete step further aggravated the situation. Model group, further thinning the thickness of the myelin sheath, the myelin lamellar discrete part is very serious, myelin formation of large balls. We can see:Compared with the sham group, the other five group are significantly different (P<0.05). There is differemce between mecobalamin group and BYHWD high-dose group(P<0.05).7.NGF secretion:More NGF immunoreactive particles were seen in sham group, BYHWD groups mecobalamin group, coloring brown. The model group NGF reduces the number of positive particles, coloring pale. In the comparison with the model group,BYHWD doses and mecobalamin group make statistically significant (P<0.01 or P<0.05)8.NGF gene expression:NGF gene expression of sham group, mecobalamin group and BYHWD groups increased significantly (4.79 times,2.16 times,3.41 times,1.83 times,1.31 times, to model group. NGF-mRNA expression of Buyanghuanwu decoction medium-dose group,high-dose group, mecobalamin group was respectively stronger than the model group (P <0.05). At the same time there is a big difference between BYHWD high-dose group and mecobalamin group (P<0.05)Conclusion1.BYHWD can effectively promote nerve crush in rats and its target organ morphology and function of skeletal muscle recovery, movement disorders of peripheral nerve damage caused by a better preventive effect.2.BYHWD has protective effect of peripheral nerve regeneration of NGF and its protein expression and the common peroneal nerve in NGF-mRNA related. The mechanism may be related to the party Dabu strength, blood circulation effect linked. The party can effectively promote the growth of axons and myelin to promote peripheral nerve regeneration, the mechanism needs further study. |
