Objective:The purpose of this study was to demonstrate the correlation between p-PDK1 and inflammatory response of SHR,and to investigate the effects of compound Uncaria antihypertension tablet on p-PDK1,ICAM-1and VCAM-1 in spontaneously hypertensive rats.Methods:Will be 24 SHR were randomly divided into three groups,each group of eight,model group,group B,he sand temple tablet group,group C,compound uncaria tablet group is group D,the other 8 with WKY rats for blank control group(group A).The experimental rats were lavage 1 times A day,eight weeks in A row,group B and group A with distilled water to fill the stomach;Group D: use a concentration of 75 mg/m L compound uncaria antihypertension tablet to fill the stomach;Group C: use a concentration of1.5 mg/ml of bei sha temple pieces to fill the stomach.Lavage in blood pressure,fixed time measuring 1 times a week during and immediately after measuring blood pressure measuring weight,according to the weight adjust dose lavage.Tail blood pressure was used to observe the blood pressure before,4 and 8weeks after the experiment,7 days a week in average 8:00~15:00 between groups of rats blood pressure measurement,for this week's blood pressure value,statistics,discusses compound uncaria antihypertensive effect on SHR blood pressure.Lavage materials after eight weeks,blood serum separation and thoracic aorta,using ELISA method to detect the rat serum ICAM-1 and the concentration of VCAM-1;Using Western Blot detection of p-PDK1 in thoracic aorta expression level.Further explore compound uncaria antihypertension tablet on serum inflammatory cytokines and VCAM-1ICAM-1,and the compound uncaria antihypertension tablet affects the quantity of p-PDK1 protein expression.In order to further investigate whether the compound uncaria step-down piece through regulating PDK1 signaling pathways to influence the SHR blood pressure to reduce inflammation reaction.Results:(1)the SHR blood pressure was significantly higher than that of WKY rats blood pressure;He sand temple tablet group and compound uncaria tablet group of SHR blood pressure decreased significantly after the treatment,in4 weeks and there has been a significant difference statistically significant.(2)SHR serum ICAM-1 and concentration of VCAM-1 was obviously higher than that of WKY rats.He sand temple tablet group and compound uncaria tablet group after treatment and VCAM-1 ICAM-1 of concentration decreased significantly.(3)the expression of p-PDK1 in SHR thoracic aorta quantity is obviously higher than that of WKY rats;He sand jotham and compound uncaria step-down pills after lavage treatment thoracic aorta tissue expression of p-PDK1 quantity significantly reduced.Conclusion:(1)compound uncaria antihypertension tablet can effectively reduce the SHR blood pressure.(2)compound uncaria antihypertension tablet can make SHR serum ICAM-1and VCAM-1 concentration decreased significantly.(3)compound uncaria antihypertension tablet can inhibit the expression of p-PDK1 in SHR thoracic aorta.Above all can draw the following conclusion:the compound uncaria antihypertension tablet may through regulating PDK1 signaling pathways that inhibit the expression of proinflammatory factor to achieve antihypertensive effect.
