Objective:We establish rat model of obesity. Observing changes in the structure of vascular intimal artery in obese rats. We study the relationship serum adiponectin levels and obesity in rat body weight, blood lipid levels, vascular endothelial nuclear factor-kappa B(NF-kB),Vascular cell adhesion molecule-1(VCAM-1),Intracellular adhesion molecule-1(ICAM-1), as well as NF-kB, VCAM-1, ICAM-1 in the expression of vascular endothelial arterial intima leading to structural changes in the mechanism.Methods:Forty eigth four-week-old healthy male Spraque-Dawley (SD) rats, rats were randomly divide into control group (18), and high-fat group (30), given high fat diet for preparating model of childhood obesity.At the end of 8 weeks,the weight of high-fat rats'body weight surpasses than in control rats to 20% of the average weight to take the obese group. Celiac artery taking blood is about 6ml,centrifugation,take the upper serum.Detect serum adiponectin levels by enzyme-linked immunosorbent assay. Detect serum total cholesterol(TC),triglyceride(TG),low-density lipoprotein cholesterol(LDL-C) and high-density lipoprotein cholesterol(HDL-C) etc, by Hitachi 7600-110 automatic biochemical analyzer. Quickly removed the heart,liver,kidney and abdominal fat(mesenteric fat and renal weeks, epididymal fat pad adjacent).Record wet weight of these organs.Free thoracoab dominal aortic length,stripping the outer membrane of adipose tissue, than taking about 2-3cm thoracoabdominal aortic aneurysm. Observing the rat intra-abdominal fat cells, liver cell morphology and structural changes in arterial intima by HE staining. The obtained empirical datum uses the SPSS 13.0 software to carry on the analysis. Results:1. High-fat group compared with the control group of SD rats gained significantly, to 4 weeks to reach statistical significance (P<0.01). At the end of 8 weeks, according to obesity standards, there were 22 rats became obese model in the experimental group.2. The obese rats organs real weight, body mass index (Lee's index), as well as visceral fat content were significantly higher than the control group (P<0.01) with statistical significance.3. Serum adiponectin levels of obese rats were(0.61±0.13) ng/ml significantly lower than the control group(1.00±0.17)ng/ml (P<0.01) with statistical significance.4. The obese rats TC, TG, LDL-C were significantly higher (P<0.01), HDL-C was significantly lower than the control group (P<0.01) with statistical significance.5. The obese rats serum adiponectin (0.61±0.13)ng/ml was negatively correlated with body weight (459.36±16.19)g and abdominal fat weight (23.65±4.22)g, (r=-0.958,-0.818, P<0.01). The obese rats serum adiponectin and TC, TG, LDL-C were significantly negatively correlated (r=-0.909,-0.930,-0.872, P<0.01), and HDL-C was positively correlated (r=0.936, P<0.01), with statistical significance.6. The number of fat cells in obese group was significantly redundant in normal group and the fat cell volume was significantly greater than the normal group at the same high-powered light microscope.Obese rats, blood vessel wall thickening,intimal smooth muscle cells move to intima, form a fatty streak or fibrous plaque.Under the naked eye liver volume obviously increases.The color assumes light yellow,has the adhesion with the peritonsillar tissue, the section greasy feeling.The liver cells of obese rats showed obvious fatty change,and hepatic lobule structure disorder in the optical microscope.7. The rat vascular endothelial NF-kB,VCAM-1,ICAM-1 were express-ed in the two groups. The obese group of vascular endothelial NF-kB, VCAM-1,ICAM-1 expression was significantly higher than the normal group (P<0.001). And obese rats of adiponectin levels and NF-kB, VCAM-1, ICAM-1 positive rate were negatively correlated(r=-0.938,-0.898,-0.891, P<0.001).Artery intima NF-kB expression and VCAM-1,ICAM-1 expression showed a significant positive correlation (r=0.662,0.671; P<0.05), with statistical significance.Conclusion:1. High fat diet can induce obesity in rats. Adipose tissue of obese rats increase in the number of fat cells. fat cells increase in volume. Liver cells fatty change. As well as artery tunica intima structural change.2. High fat diet may create organism fat metabolic disorder and causes TC, TG, LDL-C to elevate, HDL-C reduces. The abdomen internal organs Fatty tissue piles up.3. Adiponectin level of obese rat reduce, and is closely related with the fat metabolic disorder. Expected to increase serum adiponectin through the adjustment lipid.4. We know that the level reducing of adiponectin can increase ex-pression of NF-kB, VCAM-1, ICAM-1 in obese rat vascular endothelial. So that arterial intimal thickening, vascular structural changes lead to vascular diseases.
