| Objective:Study of Jiangzhi xiaobantablet is able to reduce AS TNF-alpha,IL-6plaque and the expression of PI3-K,by reducing the inflammatory reaction of AS plaque,then make AS plaques more stable,slow down the process of AS,to prevent the occurrence of ACS,to provide reliable experimental basis for the clinical prevention and treatment of AS.Methods:The establishment of atherosclerosis in New Zealand white rabbits(AS plaque)model,40 New Zealand albino rabbits,male and female each 20 rats were randomly divided into 4 groups,10 rats in each group: A.control group B.hyperlipidemia model group C.Jiangzhi xiaobantablet group D.simvastatin group.Except the A group,B group,C group and D group were given daily on the afternoon of each high fat diet feeding 50 g,100g high fat diet containing 20 mg propylthiouracil(the calculation method by 10mg-kg-1-D-1),for 3 months,to cause the prey animal atherosclerosis(plaque shape AS a)model.The clinical dosage is calculated according to the adult 70 kg,which is converted to the surface area of the moving object.Simvastatin group daily intragastric 1.96mg-kg-1-D-1;Jiangzhi xiaobantablet group gavage 0.39g-kg-1-D-1(equivalent to 2G crude drug-kg-1-D-1),A group and B group were administrated intragastrically with saline 20 m L /day groups were intragastric gavage daily for a month,time is morning to eat before.In the 3 months after the first day of each experiment in the animal after 12 hours of fasting venous blood 8-9ml from rabbit heart,sent to the laboratory were detected in all experimental animal serum lipid indicators,such as total cholesterol(TC),glycerin three greases(TG),high density lipoprotein(HDL)and low density lipoprotein(LDL)content,and the other biochemical indicators in the automatic biochemical analyzer in laboratory.4 months later,the rats were sacrificed,and the aortic arch was removed by 10% Formaldehyde Solution.After routine paraffin section,HE staining was used to examine the morphology of aortic wall.Immunohistochemical staining was used to detect the expression levels of IL-6 and TNF-.The expression level of phosphorylated P13-K was detected by immunofluorescence assay.Statistical analysis used the SPSS-22.0 medicalstatistical software package for data,all the normal examination of the main statistical indicators,statistical indexes of normal distribution are expressed as the mean + SD.(),the measurement data first variance inspection and normal distribution,if the variance is normal,the distribution,analysis of variance was used to compare between groups(post hon.by one-way ANOVA),two variable correlation analysis by linear regression(simple liner regression).Results:1,Jiangzhi xiaobantablet lipid results: B group,C group and D group before treatment,blood lipid indexes of experimental animal serum were more than the normal value,and TC,TG and LDL-C levels of the three groups of experimental animal serum significantly more than group A P<0.01).After 4 weeks in the experimental animal through the treatment group C(Jiangzhi xiaobantablet)and group D(simvastatin group)in experimental animal serum T C,TG and LDL-C levels were significantly lower than B group(model group)(P<0.01),but the A group,B group,HDL-C in C group and D group of experimental animal serum in before and after treatment there was no significant difference(P>0.05),C group and D group had no obvious difference between the two groups(P>0.05),which shows that simvastatin,Jiangzhi Xiaoban tablet can reduce serum TC,TG and LDL-C content.2,Jiangzhi xiaobantablet on expression of TNF-and AS alpha IL-6 plaque: B group(Fig.1b,2b)C group(Fig.1C,2C)and D group(Fig.1D,2D)are IL-6,TNF-alpha immunity group Brown stained positive cells expression signals and granular staining.The immunohistochemical positive cells and granular staining expression signal mainly exists in the AS plaque area.However,there was no positive expression of IL-6 and TNF-(1a,2a)in each layer of aorta in group A.B group(model group)and group A(PBS instead)between IL-6,the average gray value of IL-6 and TNF-alpha TNF-alpha,were significantly different(P<0.01),D group and B group(model group)between IL-6 and TNF-alpha IL-6,alpha TNF-average gray value comparison,significant differences(P<0.01).C group and B group(model group)between IL-6 and TNF-alpha average gray value comparison,significant differences(P<0.01),which can explain the content,effect of simvastatin Jiangzhi xiaobantablet can reduce IL-6 and TNF-immunohistochemical expression of alpha 3,Jiangzhi xiaobantablet on PI3 K protein expression in each group: experimental animal in the 4weeks after the treatment,the results showed that B group(Figure 3C),C group(Figure 3D)and D group(Figure 3B)in fluorescence microscopy by stained brown green positive expression of PI3 K protein signal,the positive expression of PI3 K protein mainly exists in the AS plaque area,but the A group There was no positive expression of PI3 K protein(Figure 3a).Moreover,the positive expression of PI3 K protein in group C(Figure 3D)and group D(Figure 3B)was significantly lower than that in group B(model group),which could be concluded that simvastatin and lipid lowering plaque could decrease the expression of PI3 K protein.Conclusion:1.The level of TNF-can decrease the expression level of the positive cells in the serum.2.The lipid lowering plaque can reduce the expression level of IL-6 immunohistochemical positive cells.3.The lipid lowering plaque can reduce the expression of PI3-K signaling pathway in atherosclerotic plaque of aorta.4.It is suggested that the plaque can reduce the inflammatory reaction in AS plaque,and make the AS plaque more stable,slow down the process of AS,prevent the occurrence of ACS,and provide a reliable experimental basis for clinical prevention and treatment of AS. |
