ABSTRACTObjective:To investigate effects of Shanhe Jiangzhi Decoction (SHJZD), a traditional Chinese medicinal prescription, on acute viral hepatitis in mouse and its mechanism. Methods: Acute liver injury was induced in mice by a single intraperitoneal injection of 0.1 mI/lOg body weight 0.12% carbon tetrachloride peanut oil solution, this model can simulate acute viral hepatitis. Mice were randomly divided into (1) normal group; (2) sham liver injury group; (3) liver injury group; (4) physiological saline+liver injury group; (5)SHJZD(lower dose)+ liver injury group; (6) SHJZD(moderate dose)+ liver injury group; (7)SHJZD(higher dose)+ liver injury group. Mice in (2)group were intraperitoneally injected peanut oil at a dose of 0.1 mi/log body weight; mice in (3) group were intraperitoneally injected 0.12% carbon tetrachloride peanut oil solution at a dose of 0.1 mlIlOg body weight; mice in (4) group had been intragastrically administered physiological saline at a dose of 0.1 ml/lOg body weight for 5 days; mice in (5).. (6) and (7) groups had been intragastrically administered SHJZD at a dose of 0.1 ml/lOg.. 0.15 mI/lOg.. 0.2 ml/lOg body weight for 5 days respectively. 0.12% carbon tetrachloride peanut oil solution were intraperitoneally injected to mice at a dose of 0.1 mI/lOg body weight, 30 mm after the latest intragastric injection of SHJZD or physiological saline. After mice were fasted for 16h, animals were sacrificed. The severity of liver injury was assessed by histological. ultrastruetrural examination, by changes in GPT and GOT in the sera and by changes in 5?Nase~ ALPase and ATPase in liver sections. Statistical analysis was2carried by ANOVA and ~ test, p < 0.05 was considered significant. Results: (1)SHJZD significantly ameliorate centrilobular necrosis in a dose-dependent manner which was demonstrated by the hepatocellualr ultrastructure; (2)SHJZD markedly decreased the activities of serum transaminases which is indexes of liver cell injury; (3)The activities of 5?Nase.. ALPase and ATPase decreased aroud central vein and portal area. Conclusion: (1)SHJZD protected the liver from CC14-induced injury as judged by morphological and biochemical observations. (2)SHJZD alleviated the leakage of 5?Nase in hepatocellualr membrane and lowered the activities of ALPase and ATPase around portal area.This may be the mechanism of SHJZD prevented against liver injury.
