| Objective:The purpose of this study was to observe the effects of the Second Pingfei Decoction(SPFD) on body weight, adipocyte, insulin resistance, hypothalamic phosphatidylinositol- 3 kinase p85 subunit(PI3K-p85), insulin receptor substrate 1(IRS1), and insulin receptor substrate 2(IRS2), protein tyrosine phosphatase 1B(PTP1B)and proopiomelanocortin(POMC) in phlegm damp type obesity rats, in order to probe the molecular mechanism of SPFD on anti-obesity research.Methods: 50 SD male rats were randomly divided into low fat diet(LFD) group(n=10) and high fat diet group(n=40). When the DIO model were established in high fat diet group, the successful model rats were randomly divided into high fat diet(HFD) group, Orlistat(OLST) group and SPFD. LFD and HFD were treated with distilled water by gavage. OLSTand SPFD were treated with orlistat(32.4mg/kg/d)and the Second Pingfei Decoction(2.7g/kg/d) by gavage respectively. After 28 days,all rats were sacrificed to detect the adipocytes by HE staining, fasting plasma glucose(FPG) by routine laboratory procedures, fasting insulin(FINS) by radioimmunoassay,PTP1 B, PI3K-p85,IRS1 and IRS2 by western blot, and POMC by RT-q PCR.Results: OLST showed watery stools, a decrease in activity, mental fatigue and yellow hairs. But SPFD had regular stools and sleek coat.The weight gain, size of adipocytes, PTP1 B and PI3K-p85 of OLST and SPFD were significant lower than HFD(P<0.01). Compared with HFD, the expression of IRS1, IRS2 and POMC in SPFD was increased. But there were no significant difference in between SPFD and HFD in FPG, FINS and HOMA-IR.Conclusion: The Second Pingfei Decoction has significant effects on weight loss. It plays its role through decreasing diameter of adipocyte, inhibiting the expression of PTP1B、PI3K-p85 and increasing IRS1, IRS2 and POMC m RNA expression. The Second Pingfei Decoction seems to be superior to Orlistat as anti-obesity therapy with less side effects such as watery stools and yellow hairs. |
