Intervention Of Pingchen Decoction On Obesity Rats With Spleen Deficiency And Phlegm Dampness Type

Objective: To observe the effect of Pingchen decoction on obese rats with spleen deficiency and phlegm dampness,and to provide new ideas and methods for the prevention and treatment of obesity in sub-health.Methods: Forty-five SPF male SD rats were randomly divided into normal control group(NC group,n=7)and model group(n=38).All rats were fed in cages in the animal room and fed freely.The obese rat model of spleen-deficiency and phlegm-dampness type was established by high-fat feeding plus nourishing the spleen,cold-dampness the spleen and long-lived wetland.The weight,general condition and spleen-deficiency integral were used to evaluate the model.Thirty-five successful rats were obtained.The rats were randomly divided into model control group(MC group,n=7),low dose intervention group(LDG group,0.5 ml/100 g,n=7),medium dose intervention group(MDG group,1 ml/100 g,n=7),high dose intervention group(HDG group,2 ml/100 g,n=7)and control group(Cel group,1.0 mg/kg/d,n=7).After4 weeks of drug intervention,blood was collected and liver and kidney function indexes [ALT,creatinine(CREA),urea(UREA)] and blood lipid indexes [lipoprotein alpha(LP-a),total cholesterol(TC),triglyceride(TG),high density lipoprotein cholesterol(HDL-C)and low density lipoprotein(LDL-C)] were detected.Liver of rats was prepared for pathological section,HE staining and optical microscopy.Histopathological changes,body mass increment and fat body ratio of rats in each group were calculated.Results:1.General situation:Compared with the normal group,the model control group showed obesity,angry bow back,greasy yellowing coat,crouching,wet and soft stool,and distinct odor in cage.Compared with model control group,Pingchen decoction treatment group improved in different degrees.LDG group rats were round,sleepy,fluffy and yellowish,curledup,soft stool,small odor in cage;MDG group rats were slightly fat,tired,fluffy white,normal stool,small odor in cage;HDG group rats were normal in shape,body spirit,two stools and coat.Often,there is almost no odor in the cage.The general state of HDG group rats was almost the same as that of normal control group rats.Compared with the model control group,the rats in Cel group were slightly fat,normal in body and mind,with white coat,general gloss,piling up and almost no odor in cage.Compared with the Pingchen decoction group,the rats in Cel group were generally better than those in LDG group and MDG group,and less than those in HDG group.2.General indicators:(1)Weight change Compared with NC group,the weight of rats in MC group increased significantly(P <0.05).After administration,the body weight of each group increased slightly,and then the weight of HDG group and MDG group began to lose,and the weight of HDG group decreased more,the weight of LDG group slowed down or maintained basically;the weight of Cel group began to lose significantly after administration,but rebounded slightly after 2 weeks;at the end of administration,LDG group,MDG group,HDG group and Cel group began to lose weight.The weight of group A was lighter than that of group MC(P < 0.05).(2)Change of integral of spleen deficiency Compared with NC group,the spleen deficiency score of model group increased gradually with the intervention process.After administration,compared with MC group,the spleen deficiency score of LDG group,MDG group,HDG group and Cel group began to decrease gradually,with the largest decrease in HDG group and the smallest in LDG group.By the end of administration,the spleen deficiency score of HDG group was close to NC group,and compared with MC group,MDG group began to decrease gradually.The scores of spleen deficiency in Cel group were also significantly lower than those in Cel group(P < 0.05).(3)Change of body mass increment Compared with NC group(P > 0.05),the increments of LDG group,MDG group,HDG group and Cel group were significantly lower than those of MC group(P <0.01),especially HDG group.(4)Change of liposome ratio Compared with NC group,the fat body ratio of MC group was significantly higher than that of NC group(P < 0.01),indicating that the fat body ratio of MC group was higher than that of normal rats;compared with MC group,the fat body ratio of LDG group,MDG group,HDG group and Cel group was smaller than that of MC group(P< 0.05),and the fat body ratio of HDG group was almost the same as that of NC group.3.Biochemical indicators:(1)Changes of blood lipid Compared with NC group,the blood lipid in MC group increased significantly(P <0.01);compared with MC group,the blood lipid in LDG group,MDG group,HDG group and Cel group decreased in varying degrees(P < 0.05).The results of LP-a showed that there was no significant difference among the groups(P > 0.05).(2)Changes of liver and kidney function Compared with NC group,ALT in MC group increased significantly(P < 0.01).Compared with MC group,ALT in LDG group,MDG group,HDG group and Cel group was lower(P < 0.01).There was no significant difference between CREA and UREA groups(P > 0.05).4.Liver pathology:Nucleus morphology of NC group was regular,and no obvious vacuolar lipid droplets were observed in MC group;a large number of vacuolar lipid droplets were observed in MC group;liver pathological morphology of LDG group,MDG group,HDG group and Cel group was improved in varying degrees compared with MC group,especially HDG group,which had a significant effect,similar to NC group.Conclusion:1.The obese rat model of spleen-deficiency and phlegm-dampness type can be successfully established by multi-factor intervention method of high-fat feeding plus nourishing the spleen,cold-dampness the spleen and long-lived wetland.2.Pingchen Decoction can reduce the weight of obese rats with spleen deficiency and phlegm dampness and improve the obesity status.Its effect is related to the dosage,and the effect of high dosage is better.Compared with Tripterygium wilfordii,Pingchen Decoction has a more significant effect in high dosage.
