Oedaleus infernalis Sauss belongs to Oedipodidae in Orthoptera, It is a common insect which can be found in most regions of China. The current studies of Orthoptera were most focused on analysing its morphological, genetic diversity and mitochondrial genome. There are few researches about miRNA and transtcriptome comparehensively in Oedaleus infernalis Sauss.The main idea of this experiment was to build three different libraries of miRNA and transcriptomes of male adult, female adult and nymph to further examine the sequences via Illumina HiSeq 2000 high-throughput sequencing platform. What is more, The bioinformatics methods were used to annotated the the differentially expressed genes, analysing GO function and KEGG metabolic pathway. And also the known miRNA and novel potential miRNA candidates. The main conclusions are as follows:(1) The male libraries obtained a total of 947 miRNA, the female libraries gained 873 miRNA and 904 miRNA were found in the nymphs'libraries, with 3,549, 697?32,61,224 and 3,316,811 reads respectively.(2) Using miRDeep we found that the novel miRNAs of male, female and nymph were predicted in this experiment found 30 novel miRNA for male,43 for female and 32 for nymphs.(3) We analysised the differential expressed miRNA pattern of male and female adults libraries, and discovered 112 siganificantly differential miRNAs and 52 novel miRNA. In the miRNA expression pattern of adult and nymph libraries, there are 499 conserved miRNAs were siganificantly differentially expressed and 44 novel miRNA were found. A total of 403 target genes were predicted in differentially expressed miRNAs of male and female, and 516 target genes were predicted in adult and nymphs.(4) The transcriptomic secquencing for three samples of male, female and nymph obtained 54,971,450?52,666,682 and 53,132,988 clean reads respectively. A total of 51,308 Unigenes were obtained from the combined assemblies with an overall length of 57,606,410 nt. There are 52,743 Unigenes were found in male adult,57,437 Unigenes in female adult and 57,608 Unigenes in nymph.(5) Unigenes were annotated to Nr (22,650), Nt (12,789), Swiss-Port (18,701), KEGG (16,724), COG (9,594) and GO (10,839) databases, The total annotated unigenes were 51,608, accouting for 43.50% of All-Unigenes. These databases can be used in studying the genome and transcriptome of Oedaleus infernalis Sauss.(6) The analysis of differentially expression gene and KEGG pathway annotation of the three samples showed that the male adult expressed more genes in cellular membrane and enzyme activity, however, the gene expression in female adult were more activite in metabolism pathway for reproduction. The nymphs have more genes expressed in maintaining a normal level of body metabolism. These information of gene annotation laid the foundation for further research of Oedaleus infernalis Sauss...
