Chorthippus chinensis Tarbinsky belongs to subgenus Megaulacobolhrus, genus Chorthippus of Areypteridae in Orthoptera. Some research have done about this genus mainly based on morphology and molecular makers. However, disputes over species identification and classification remain. With High-throughput technologies becoming more sophisticated, it is possible to study genome, transtcriptome and miRNA compreh ensively.In this study transcriptome and small RNA of male and female C. chinensis were sequenced using HiseqTM2000 platform. In transcriptome analysis, we carried out functional annotation of differentially expressed genes (DEGs) with GO function and KEGG metabolic pathway to better understand the main biological functions and metabolic pathways paticipating by those DEGs. As for miRNA, we conducted categorization annotation, comparison with known miRNA, differential expression analysis, target gene prediction, novel miRNA prediction etc. The main conclusions obtained from these study are as follows:(1) A total of 47,275 unigenes obtained from combined assemblies of male and female C.chinensis with total length 45,566,140nt, mean length 964nt, and N50 1,763nt.56,447 unigenes obtained from female adult, the total length and mean length were 39,236,142nt and 695nt respectively, and N50 was 1,455nt.50,222 unigenes obtained from male adult, the total length and mean length were 35,926,168nt,715nt, respectively, and N50 was 1,459. Then male and female unigenes were assembled and redundancy removed to get 47,275 All-unigenes.(2)Unigenes which were annotated to NR, NT, Swiss-Prot, KEGG, COG and GO database were 20784,10822,17257,15183,8516 and 10544 respectively. The total annotated unigenes had been reached 22,544, accounting for 47.69% of All-unigenes.
