Mesenchymal stem cells(MSCs)have the functions of immune regulation,lineage differentiation,and secretion of cytoactive factors.Therefore,MSCs are favored by cell therapy researchers in repairing damaged tissues and treating autoimmune diseases.MSCs can be extracted from bone marrow,dental pulp,adipose tissue,peripheral blood,umbilical cord and other tissues.However,the application of MSCs needs to solve the following problems: 1.The gene expression profiles of MSCs;2.Although many media were used to culture MSCs,these different media produce these MSC cells with varied characteristics.It is unclear that the effects of serum and serum-free systems on expression profiles of MSCs;3.Effects of passaging expansion and culture methods on gene expression profiles of MSCs.Compared with other tissue-derived MSCs,human umbilical cord mesenchymal stem cells(h UCMSCs)and dental pulp stem cells(h DPSCs)are easily accessible and have high proliferation ability and low immunogenicity.Therefore,h UCMSCs and h DPSCs have attracted worldwide attentions.To address these issues,we used h UCMSCs and h DPSCs as representative MSCs to study gene expression profiles of MSCs and effects of culture conditions and different generations by transcriptome sequencing(RNA-Seq)and bioinformatics analysis.The results show that: 1.h UCMSCs are superior to h DPSCs in cell proliferation and immune regulation,and h UCMSCs have a smaller cancer risk;2.SFM serum-free system has greater advantages in cell proliferation,cell morphology maintenance and immune regulation,etc.But it is closely related to cancer progression;3.There are fewer differential genes between different generations of cells under the SFM system,but accompanying with the intensive expansion,h UCMSCs display the weaker proliferative ability and increase more cancer risk genes.This study uncovers the unique biological characteristics of h UCMSCs at the gene expression level,which provides a reference for the future application of h UCMSCs.
