Bladder cancer occurring on the bladder mucosa is the most common malignancy of the urinary system known as its high morbidity and mortality. However, many weakness and shortcomings still exist in current clinical examination detection and diagnosis for bladder cancer, such as invasive detection, low specificity and sensibility. In order to realize more accurate detection, this paper proposes a new type of microfluidic chip used for the early detection of bladder cancer, as well as the design and fabrication of the chip.In this paper, PDMS is used as the material of the microfluidic chip for production utilizing cast molding method. Bladder cancer cells are collected from patients'urine sample utilizing a three-step centrifugation and then identified by immunohistochemical staining with specific antibody. Specific antibody is fixed onto a glass slide after silanization modification and used to capture bladder cancer cells in specific cylindrical micropattem and cell capturing chamber as a result of antigen-antibody immunosorbent assay. Bladder cancer cells could be captured and identified in the chip, and then be observed under microscope with different perfusion speeds and capturing conditions.The results showed that specific antibody could be fixed onto the surface of the slide and restored for the capturing of bladder cancer cells in the presented microfluidic chip, and fluorescence intensity could be regard as the ability of slides to fix antibody with fluorescence. During the exploring experiment,5 minutes of silanization and 2 hours of antibody fixing are proved to be the best condition for antibody fixing onto slide. Since bladder cancer cells are collected from the patients'urine sample and identified through immunohistochemical method with specific antibody Anti-CD 15, and the results show that some cells in the sample are marked with fluorescence. Bladder cancer cell capture and detection experiment in the chip show that all captured cells are marked with fluorescence, which proves that the microfluidic chip could capture bladder cancer cells in the sample specifically.These findings proved that this study has successfully established the microfluidic chip applied to the detection and diagnosis of bladder cancer, and explored the best condition for antibody fixing onto the bottom of microfluidic chip, and also extracted bladder cancer cells in the patients' urine sample, as well as the detection, which provides powerful platform for early detection and diagnosis of bladder cancer.
