Cell culture technology is one of the most basic technologies in biological research.The external environment of cell culture environment must be kept constant,including suitable temperature,humidity,CO2 concentration and specific gas environment.Traditional cell culture using culture plate,culture bottle and other devices can not precisely restore the body's micro environment.Microfluidic chip has the characteristics of miniaturization,integration and multi flux.It can control the microenvironment accurately and provide new ideas and methods for cell culture.At present,microfluidic cell culture depends on cell culture box and microfluidic cell culture.When cultured in cell culture box,the cell growth state can not be monitored by microscope.It is difficult to achieve the function of monitoring while culturing,while microfluidic cell culture platform is expensive,which is not conducive to the promotion in scientific research.Therefore,the research on the measurement and control system of external environmental factors in microfluidic chip cell culture is of great significance for promoting the progress of biomedical engineering technology.On the basis of the analysis of the research status of the cell culture environment control device of microfluidic chip,the measurement and control system of external environment factor for cell culture of microfluidic chip is developed.The main contents of the work are summarized as follows:(1)According to the conditions and requirements of cell culture of microfluidic chip,and aiming at the problem that can not be realized online,a device that can get rid of the limit of the incubator and combine conventional inverted microscope to realize the on-line monitoring of cell culture is designed.(2)Aiming at the insufficient mixture of gas in the process of experiment,a micro mixer is designed to realize dynamic mixing of gas by controlling the movement path of gas.(3)In order to avoid the influence of temperature and humidity coupling on the measurement and control system,the coupling system is decomposed into two independent systems through designing the understanding of coupling algorithm by feedforward compensation.(4)The experiment platform of theexternal environment factor measurement and control system for the flow control chip cell is constructed,and the hardware platform and software platform of the detection module,control module and display module are completed.(5)Experimental verification and analysis were carried out to verify the feasibility of the measurement and control system for external environmental factors of microfluidic chip cell culture.The experimental results show that: The measurement and control system of external environmental factors designed by the paper can effectively maintain the normal growth of cells on microfluidic chips and realize the function of monitoring while training;The micro mixer device proposed in this paper can make the gas mixing more uniform and avoids the effect on normal growth of cells;Through the comparative analysis of the adaptive decoupling simulation and the PID simulation,it can be found that the problem of temperature and humidity couplingis solved.The temperature error is less than 0.5 and the control performance of the system is improved.
