With the improvement of people's living standards and enhancement of people's health consciousness, the detection of biomarkers becomes more and more important, the traditional biomarker detection methods, such as liquid chromatography-mass spectrometry, gas chrom-atography-mass spectrometry, require special operations of large-scale instruments, and suffer from disadvantages including expensive, time-consuming and so on. Microfluidic paper chip, is a new platform with many advantages such as light quality, low cost, good biocom-patibility, no contamination. This paper, take clenbuterol and cortisol as examples, achieved two detections on our two initially designed paper chips combined with specificity of immune reaction and high sensitivity of SERS. Specifically, we achieved:(1) Determination of beta-agonists by paper microfluidics coupled with SERS and immune reactionClenbuterol can promote fat decomposition, improve the lean meat rate of animal body, but is banned in Chinese meat industry for it's damage to people. To prevent the abuse of illegal use, determination of clenbuterol is necessary. We extracted the clenbuterol in swine hair and the switch-type paper microfluidic used in this study was fabricated by wax printing. 55nm Au nanoparticles are synthesized, during connection process between the MBA mole-cular and Au nanoparticles, the incubation time and volume are optimized. Based on that best conditions, SERS nanoprobe-grafted CL antibody are synthesized. different enhancement effects on different paper are measured and the Whatman No.1 filter paper chosed to be the experiment materials by the strongest signal. On the switch-type chip, we realized the filtra-tion and enrichment of swine hair extract. Finally, competitive immune reaction between clen-buterol antigen, clenbuterol antibody and samples contained clenbuterol controlled by the switches on the chip. due to three parallel units with three switches, three reactions can be achieved at the same time. the limit of detection can reach 0.1 pg/mL.(2) Determination of cortisol by paper microfluidics coupled with SERS and immune reactionCortisol is an important index to evaluate the stress level, detection of cortisol can be characterized as the objective basis of people's psychological pressure, especially detection of hair cortisol can reflect the people's long-term psychological pressure objectively. We synthe-sized 55nm Au nanoparticles, based on that best conditions above, SERS nanoprobe-grafted cortisol antibody are synthesized. the whatman NO.1 filter paper was chose to be the exper-iment materials to produce the switch-type chip, we realized the filtration and enrichment of hair extract and competitive immune reaction between cortisol antigen, cortisol antibody and samples contained cortisol.96 parallel unit helps realizing high-throughput detection, the lim-it of detection can reach 0.1 pg/mL. The detection is achieved on a portable Raman instrum-ent, too. the limit of detection can reach 1 pg/mL. laiding the foundation for the real-time detection of cortisol.
