Objective: To investigate the role of mi R-1290 in the occurrence and development of gastric cancer(GC),and provide a theoretical and experimental evidence for the diagnosis and clinical treatment of GC.And to analyze the relationship between the epression of mi R-1290 and infection of Helicobacter pylori(Hp),explore the pathogenesis mechanism of Hp-associated GC.Methods: Gastric mucosal biopsies were collected from 30 patients with GC,30 patients with chronic atrophic gastritis(CAG)and 30 patients with chronic superficial gastritis(CSG),which diagnosed by gastroscopy and gastric mucosal biopsy.The infection status of Hp was determined by Giemsa staining method and the expression level of mi R-1290 was detected by Quantitative real-time PCR(q PCR)in each group.Results: 1.The relative expression quantity of mi R-1290 in GC tissues was significantly lower than that in CAG group and CSG group(P<0.05),but there was no significant difference between CAG group and CSG group(P>0.05).2.The expression level of mi R-1290 in GC tissues was correlated with tumor differentiation,depth of invasion and TNM stage(P<0.05),but there was no statistical differences among patient's gender,age,tumor size,lymph node metastasis and distant metastasis(P>0.05).3.The relative expression quantity of mi R-1290 in Hp-positive GC,CAG and CSG gastric mucosal biopsies were lower than that of Hp-negative biopsies at the same grade lesion(P<0.05).4.The relative expression quantity of mi R-1290 in Hp-positive GC tissues was significantly lower than that in Hp-positive CAG and CSG gastric mucosa tissues(P<0.05),while the difference between Hp-positive CAG and CSG gastric mucosa tissues was not statistically significant(P>0.05).Conclusion: 1.The expression of mi R-1290 in GC tissues was significantly lower than that in CAG and CSG gastric mucosa tissues,suggested that the down-regulation of mi R-1290 expression may be related to the occurrence and development of GC.2.The expression of mi R-1290 in poorly differentiated GC tissues was significantly lower than that in moderately and highly differentiated GC.The expression of mi R-1290 in GC tissues whose infiltration depth reached stage T3 and T4 was lower than that in stage T1 and T2 GC.The expression of mi R-1290 in GC tissues whose TNM stage reached stage III and IV was lower than that in stage I and II GC.These results indicated that the low expression of mi R-1290 in gastric mucosa tissues was closely related to the differentiation,infiltration and development of GC cells.The dynamic detection of the expression of mi R-1290 in gastric mucosa tissues may provide certain reference to detecte the early GC,observe the therapeutic effect,and even judge the prognosis.3.For GC,CAG and CSG patients,the expression of mi R-1290 in Hp-positive gastric mucosa tissues was lower than that in Hp-negative tissues,suggested that the infection of Hp may inhibit the expression of mi R-1290 in gastric mucosa tissues through certain mechanisms,and then participate in the occurrence and development processes of GC.
