Objective:Looking for molecular markers in the early diagnosis of gastric cancer, further research the multifaceted pathogenesis.Methods:The subjects were divided into three groups, Helicobacter pylori positive include the control group, the gastric precancerours group, the gastri cancer group. Helicobacter pylori negative include the control group, the gastric precancerours group, the gastri cancer group. By ELISA to detect serum and gastric juice, IL-6, IL-8level changes. Using immunohistochemical method to detect gastric biopsy tissue sections of patients in each group IL-6, IL-8expression.ResultS:(1) In H.pylori positive group, the level of IL-6and IL-8in serum of patients with gastric cancer is higher than the control group, with significant difference between them (P<0.05).The level of IL-6and IL-8in serum of gastric precancerous lesions is lower than gastric cancer (P<0.05), but still higher than normal level (P<0.05).In H.Pylori-negative group, the level IL-6and IL-8in serum of patients with gastric cancer are higher than those of control group, with significant difference between them (P<0.05).IL-6and L-8serum levels in gastric precancerous disease group lower than gastric cancer group (P<0.05), but still higher than normal level (P<0.05).IL-6and IL-8level in serum in Helicobacter pylori positive group is higher than the same grade lesion in H.Pylori-negative group, the difference was significant (P<0.05).(2) In H.pylori positive group, the level of IL-6and IL-8in gastric juice of patients with gastric cancer is higher than the control group, with significant difference between them (P<0.05). The level of IL-6and IL-8in gastric juice of gastric precancerous lesions is lower than gastric cancer (P<0.05), but still higher than normal level (P<0.05). In H.Pylori-negative group, the level IL-6and IL-8in gastric juice of patients with gastric cancer are higher than those of control group, with significant difference between them (P<0.05). IL-6and L-8gastric juice levels in gastric precancerous disease group lower than gastric cancer group (P<0.05), but still higher than normal level (P<0.05). IL-6and IL-8level in serum in Helicobacter pylori positive group is higher than the same grade lesion in H.Pylori-negative group, the difference was significant (P<0.05).(3) In H. pylori-positive group, IL-6in the control group, precancerous lesions and gastric cancer positive expression rate was13.3%,38.5%and84.0%, a statistically significant difference between the groups (P<0.05). IL-8in the control group, precancerous lesions and gastric cancer patients positive expression rates were20.7%,33.3%and70.1%, difference between the groups was statistically significant (P<0.05).H. pylori-negative group, IL-6in the control group, the positive expression rates of precancerous lesions and gastric carcinoma were9.1%,33.1%and65.2%,difference between the groups was statistically significant (P<0.05). IL-8in the control group, precancerous lesions and gastric cancer patients positive expression rates were14.7%,29.3%and62.0%difference between the groups was statistically significant (P<0.05).Conclusions:(1) Detect the changes of IL-6and IL-8in the serum of patients with gastric cancer for the understanding of the immune status, understand their health condition, the degree of development of the preoperative evaluation of patients with gastric cancer tumor has important clinical reference value.(2) IL-6and IL-8in gastric juice change to some extent reflects the gastric pathophysiological state. the changes of IL-6and IL-8in gastric juice can effectively distinguish between normal and diseased gastric mucosa.(3) IL-6and IL-8were significantly higher in the cancer tissue. In gastric mucosa of IL-6and IL-8expression in gastric cancer patients to some extent reflect the changes in the progression of the disease. Detecting cancer tissue levels of IL-6and IL-8on the diagnosis and clinical treatment of patients with gastric cancer is important.(4) The IL-6and IL-8differentially expressed in the same grade lesion in H. pylori-positive and H. pylori-negative prove that H. pylori can be enhanced IL-6and IL-8expression, and promote the occurrence of gastric cancer.(5) IL-6and IL-8is a scientific and accurate indicator of gastric cancer detection and condition monitoring.
