Effects And Mechanism Of Anthricin Combined With 5-Fluorouracil On Colorectal Cancer In Vivo

Objective:To investigate the effect of Anthriscus sylvestris petroleum ether part(APE)and Anthricin on tumor-bearing mice and the mechanism including apoptosis,inflammation and adhesion.Methods:1.Balb/c mice xenotransplant model of colorectal cancer CT26 was established.The mice were treated with saline,anthricin,5-FU and the combined of anthricin with 5-FU.Then observed the change of mice and tumor.The pathological state of tumor was detected by HE staining.The expression of PI3 K,AKT,NF-?B,BCl-2 and Bax were tested using Western Blot and immunohistochemical method.2.Balb/c nude mice xenotransplant model of colorectal cancer HT29 were established.The mice were treated with anthricin,5-FU and the combination of 5-FU with low,medium and high anthricin(control group were given saline and sodium carboxymethyl cellulose).Then observed the changes of mice and tumor.The pathological state of tumor was detected by HE staining.The expression of FAK,MMP-9,iNOS and NF-?B were tested using Western Blot and immunohistochemical method.3.Balb/c nude mice xenotransplant model of colorectal cancer HT29 were established.The mice were treated with APE,5-FU and the combination of 5-FU with low,medium and high APE(control group were given saline and sodium carboxymethyl cellulose).Then observed the change of mice and tumor.The pathological state of tumor was detected by HE staining.The expression of FAK,MMP-9,iNOS and NF-?B were tested using Western Blot and immunohistochemical method.Results:1.Anthricin could enhance the anti-tumor effects of 5-FU on CT26 tumor-bearing mice.It had efficacy enhancing and toxicity reducing with 5-FU in the treatment of colorectal cancer.The treatment of anthricin with 5-FU could markedly inhibit the Bcl-2?PI3K?AKT?NF-?B expression,and promote the expression of Bax.The combination medicine probably interferes the activation of anti-apoptosis factors through PI3K/AKT signaling pathway,inducing cell apoptosis,to inhibit the tumor growth.2.Anthricin could enhance the anti-tumor effects of 5-FU on HT29 tumor-bearing nude mice,it had efficacy enhancing and toxicity reducing with 5-FU in the treatment of colorectal cancer.The treatment of anthricin with 5-FU could markedly inhibit the FAK and MMP-9 expression,the combination medicine had synergistic effect on adhesion factors,led to increase of cellular adhesion and decrease of colorectal cancer cell migration The treatment of anthricin with 5-FU could markedly inhibit the iNOS and NF-?B expression,the combination medicine may had synergistic effect to the degree in influencing inflammatory factors,led to inhibition of inflammatory factors expression,to achieve anti-inflammatory effect.3.APE could enhance the anti-tumor effects of 5-FU,it had efficacy enhancing with 5-FU in the treatment of colorectal cancer.The treatment of APE with 5-FU could markedly inhibit the FAK and MMP-9 expression,the combination medicine had synergistic effect in influencing adhesion factors,led to increase of cellular adhesion and decrease of colorectal cancer cell migration.The treatment of APE with 5-FU could markedly inhibit the iNOS and NF-?B expression,but they didn't show any synergistic effect.
