Objective:To establish a method for the analysis of anthocyanins in Berberis atrocarpa,raspberry,medicine mulberry and chrysanthemum,and to study on the antioxidant activity of anthocyanins;to identify raspberry(Rubus idaeus L.)and its confusable product raspberry(Rubuschingii Hu).Methods:1.Determination of DPPH Free radical scavenging antioxidant activity of Berberis atrocarpa,Raspberry medicine mulberry and chrysanthemum in Xinjiang by UV-vis method.2.The anthocyanins were identified qualitatively by TLC in Berberis atrocarpa and other plants,and the antioxidative active components were analyzed by TLC-bioautography.3.Determination of anthocyanins in Berberis atrocarpa,raspberry and medicine mulberry by TLC-scanning.4.The HPLC method was established for the determination of anthocyanins in raspberries and medicine mulberry.5.The raspberry(Rubus idaeus L.)and raspberry(Rubuschingii Hu)were identified by TLC and HPLC in the Pharmacopoeia,and the antioxidant activity of the two were measured.Results:1.The results of DPPH free radical scavenging test showed that the IC50 of Berberis atrocarpa,raspberry,medicine mulberry and chrysanthemum were 0.02 mg/mL,1.00 mg/mL,1.20 mg/mL and 0.16 mg/mL.2.The results of TLC showed that anthocyanins had strong antioxidant activity,There are three anthocyanins in Berberis atrocarpa,known as cyanidin-3-O-glucoside,three anthocyanins in raspberry,and two anthocyanin components in raspberry,known as cyanidin-3-O-glucoside and cyanidin-3-O-rutinoside.There are two anthocyanins in chrysanthemum,but the specific components are unknown.There are three kinds of anthocyanin components in medicine mulberry,known components are cyanidin-3-O-glucoside and cyanidin-3-O-rutinoside.3.A method for the determination of Berberis atrocarpa.raspberry and medicine mulberry based on TLC-scanning was established.The content of cyanidin-3-O-glucoside was 1.7265 mg/g in Berberis atrocarpa,the content of cyanidin-3-O-glucoside in raspberry was 0.34 mg/g and the content of cyanidin-3-O-rutinoside in raspberry was 0.29 mg/g;the content of cyanidin-3-O-glucoside and cyanidin-3-O-rutinoside in medicine mulberry was 0.34 mg/g and 0.29 mg/g,respectively.The contents of cyanidin-3-O-rutinoside were 2.49 mg/g and 1.50 mg/g,respectively.4.A HPLC method for the determination of anthocyanins in Berberis atrocarpa,raspberries and medicine mulberry was established.The content of cyanidin-3-O-glucoside was 1.94 mg/g in Berberis atrocarpa,and the contents of cyanidin-3-O-glucoside and cyanidin-3-O-rutinoside were 2.94 mg/g and 0.92 mg/g,respectively;The contents of cyanidin-3-O-glucoside in two batches of raspberries were 0.874 mg/g and 0.867 mg/g,respectively.The contents of cyanidin-3-O-rutinoside in two batches of raspberry were 0.544 mg/g and 0.604 mg/g by external standard method,and the contents of cyanidin-3-O-rutinoside in two batches of raspberry were 0.564 mg/g and 0.687 mg/g by a test of multiple assessment methods,respectively.5.The results of TLC and HPLC showed that the raspberry(Rubuschingii Hu)contains kaempferol-3-O-rutinoside,linden glycoside and ellagic acid,while raspberry(Rubus idaeus L.)contains only a small amount of ellagic acid,mainly anthocyanin.The results of DPPH free radical scavenging experiment showed that both raspberry(Rubus idaeus L.)and raspberry(Rubuschingii Hu)could scavenge DPPH.Conclusion:1.Berberis atrocarpa,raspberry,medicine mulberry and chrysanthemum all have the ability of scavenging DPPH,and the anthocyanins have antioxidant activity.2.The results of TLC-scanning and HPLC content determination met the requirements.Under the condition of the lack of cyanidin-3-O-rutinoside reference substance and the high cost,the simultaneous determination of multi-components of medicinal materials could be realized by a multi-evaluation method.3.Raspberries(Rubus idaeus L.)and raspberries(Rubuschingii Hu)belong to two species of the same family,raspberry(Rubus idaeus L.)and raspberry(Rubuschingii Hu)are similar in appearance,but their main chemical components are different.
