| Blackberry Marc is the waste in the food industry after extracting juice. Studies show that the BlackBerry marc is rich with polyphenols, and if we can utilize it fully, we will receive good value from it. In this paper we screen antioxidant activity fraction through comprehensive evaluation of the antioxidant activity from BlackBerry marc and study such as Optimum extraction process to provide scientific basis of BlackBerry resources better use. The main contents include about:The blackberry pomaces was vibration extracted using different solvent including aqueous, aqueous (pH3),60% ethanol,80% ethanol,95%ethanol, n-butane and ethyl acetate in room temperature respectively. And then the different extract was obtained. The total·OH antioxidant capacity and O2-·and DPPH·Free radical scavenging capacity of the different extracts was determined spectrophoto metrically using the total antioxidant capacity assay system, DPPH·system, hydroxyl radical system, superoxide radical system. The results show that all the extracts from the blackberry pomaces have antioxidant activity and free radical scavenging activity. The acidic solvent has better effect than non-acidic solvent. The 60% ethanol is the appropriate solvent. And the total antioxidant capacity, scavenging capacity to O2-·and DPPH·of 60%ethanol extract all are weaker than Vc, but the scavenging capacity to·OH is stronger than Vc. DPPH is combined with the HPLC method to evaluation the radical scavenging capacity (RSC) and the results show that the DPPH-HPLC method can be successfully applied to measure the free radical-scavenging activity of blackberry pomaces, Meanwhile, the DPPH-HPLC method is superior to colorimetric because there is no interference from color pigments.The anthocyanin in the blackberry pomace was extracted with the different methods and the results showed that the anthocyanin content of resound-assisted extraction is the highest. The 60% ethanol was used to extract anthocyanin with ultrasonic assisted extraction and ratio of liquid to material, ultrasonic time, extraction temperature, ultrasonic power, extraction times were assessed in single-factors experiment. In basis of single-factor test, the optimum conditions for the ultrasound extraction of anthocyanin from blackberry pomaces are determined using response surface method(RSM). The results show that the anthocyanin ultrasonic assisted extraction conditions with 60% ethanol are:the liquid-solid ratio(ml:g) 14:1, extraction time 40min, extraction temperature 58℃, ultrasonic wave power 300W, two step extraction.Correlation analysis showed that there was significant correlation between anthocyanin content and DPPH·scavenging activity, and inferenced that anthocyanin is the main free radical scavenging substances. |
