Anthocyanins is one kind of water-soluble natural pigment, which widely exists in the flavone compounds, mainly exists in the form of glycoside, so it is called anthocyanins. The anthocyanins are the most effective natural water-soluble free radical scavenger so far. As research continues, the effect of anthocyanins has attracted the attention of the researchers, the main functions include anti-aging, activate the macrophage, make serum immunoglobulin from free radical, enhance human immunity, protect blood vessels, reduce the incidence of cardiovascular disease, reduce the risk of diabetes and complication occurrence probability, improve the degenerative dementia, protect genes in cells from attack, reduce the incidence of various cancers, anti-inflammatory, prevention, visually impaired, and many other functions.Blueberry has realized industrialization cultivation in china, which has nearly50000units of area of cultivated area, southern Liaoning province has nearly10000units of area of cultivated area; In the next few years, blueberry fruit will enter besiktas,40%of blueberry fruit was consumed as fresh fruit,60%of blueberry fruit was processed. At present, the quality of blueberry fproducts on the market are good and bad intermingled, with no industry standards, the quality of the products and the establishment of the evaluation standard are imminent.Blueberry anthocyanin’s research focuses on the separation, purification and identification. This paper carried out based on the early stage of the anthocyanins detection, high performance liquid chromatography (HPLC) for the main cultivated varieties of anthocyanin composition and content of southern region analysis, at the same time, the evaluation activity of extracts of blueberry anthocyanin scavenging free radicals, and at the cellular level to investigate antioxidant activity, establish a blueberries’s product quality and evaluation standard, and get the following conclusion:(1) Different varieties of blueberry anthocyanin content determinationChoose the black pearl, north green, berkeley, bluecrop abundant four blueberry varieties, high performance liquid chromatography (HPLC) analysis of the fresh fruit, found13kinds of main anthocyanins in blue berries, the13kinds of anthocyanins in black pearl, Berkeley,north green, the proportion of total anthocyanins in Berkeley were89.63%,99.32%,99.04%,98.26%.In the13kinds of anthocyanins as a fingerprint of blueberry anthocyanin components, as a blueberry anthocyanin qualitative and quantitative analysis of evaluation criteria;Knapweed pigment as standard, the determination of the black pearl, north green, berkeley, bluecrop anthocyanin varieties and content respectively, anthocyanin content comparison results for the black pearl>north green>berkeley>bluecrop, anthocyanin anthocyanins content in sample was30.45%, in addition, for the five varieties, respectively, to determine the DPPH radical scavenging capacity, and comparing them to draw black pearl varieties blueberry anthocyanin content high, DPPH radical scavenging ability is strongest.(2) The blueberry anthocyanin activity of scavenging free radicalsWith vitaminC (VC), antioxidant2,6-2tertiary butyl-4-methyl phenol butylated hydroxytoluene (BHT) as the contrast, with DPPH free radicals, ABTS free radicals, superoxide anion, hydroxyl free radical removal ability, total antioxidant activity, reducing power and inhibition of lipid peroxidation as an index, respectively blueberry anthocyanin extraction ability of scavenging free radicals. Experiments show that in addition to the inhibition of lipid peroxidation, blueberry anthocyanin extracts (anthocyanin content of30.45%) ability of scavenging free radicals is proportional to the content of anthocyanin, on DPPH free radicals, superoxide anion, hydroxyl free radical removal ability, total antioxidant capacity, anthocyanins are stronger than VC unit mass activity, quality of ABTS radical scavenging capacity of the unit active anthocyanin and VC. Inhibition of lipid peroxidation, fat-soluble butylated hydroxytoluene stronger than the blueberry anthocyanin and VC.Black pearl, north green, berkeley, bluecrop abundant four varieties of blueberry anthocyanin content and DPPH free radicals were positively correlated.(3) The blueberry anthocyanin cellular level antioxidant activityBGC-823as the object, through add cell oxidative damage induced by H2O2, blueberry anthocyanin extract, VC and butylated hydroxytoluene effect on protection and restoration of damaged cells, shows obvious effect on protection of the blueberry anthocyanin extracts, and has a dose dependent. Blueberry anthocyanin extract, VC and butylated hydroxytoluene on H2O2cells were not recover after injury. Adding H2O2induced BGC-823cells increase ROS, and blueberry anthocyanin add inhibits the generation of ROS, and a dose of positive correlation. At the same time, can improve the intracellular glutathione peroxidase, catalase activity, removal of oxygen free radicals.In conclusion, the paper confirmed the13kinds of anthocyanins as blueberry anthocyanin fingerprint components, used for qualitative and quantitative analysis of HPLC. Blueberry anthocyanin removal ability is proportional to the content, the DPPH free radicals, superoxide anion, hydroxyl free radical removal ability, total antioxidant capacity, unit mass activity anthocyanin is better than that of VC, while the ABTS radical scavenging capacity and VC. Blueberry anthocyanin, at a cellular level can be increased by inhibiting ROS, increase the cellular glutathione peroxidase, catalase activity, removal of oxygen free radicals, the protection of cells.
