Lipid deposition is the accumulation of lipid droplets in liver,pancreas or adipose tissue.Lipid deposition has certain lipid toxicity on skeletal muscle,pancreatic ?-cells,liver and so on,and it is closely related with diabetes,fatty liver,hyperlipidemia and other diseases.The existing lipid-lowering drugs have some side effects and adverse reactions.Cherry is rich in various active nutrients,including anthocyanin content of up to 4-450mg/100g fresh weight.Cherry anthocyanins have antioxidant,prevent cardiovascular disease and lipid-lowering and other activity,it can reduce cholesterol(TC),triglyceride(TG),low-density lipoprotein cholesterol(LDL-C)levels,increase apolipoprotein(ApoA)and high-density lipoprotein-cholesterol(HDL-C).Therefore,separation and preparation of anthocyanin from cherry and study its lipid-lowering effect is of great significance.In this paper,the contents of total flavonoids and total polyphenols in cherry were determined and the antioxidation activity of cherry anthocyanins and its inhibition on cell lipid deposition were studied.The role of cell autophagy in cherry anthocyanin inhibition of cell lipid deposition were also studied.The main results are as follows:1.The main components of cyanine anthocyanins were cyanidin-3-,cyanidin-3-rutinoside and procyanidin-3-glucoside,which were identified by HPLC.Antioxidant activity of cherry anthocyanins was evaluated by the in vitro antioxidant test of DPPH and ATBS and cell experiment combining with fluorescence probe.The results showed that the antioxidant activity of cherry anthocyanins was strong in vitro and had a better protective effect of cell oxidative stress model.2.The cell fat deposition model induced by oleic acid(OA)induced by HepG2 and L02 cells was used to evaluate the inhibitory effect of cherry anthocyanins on the inhibition of cell lipid deposition.It was confirmed that cherry anthocyanin significantly improved the lipid deposition of HepG2 and L02 cells.The levels of triglyceride(TG),total cholesterol(TC),alanine aminotransferase(ALT)and aspartate aminotransferase(AST)in the cells were significantly decreased.3.Combined with TEM and immunofluorescence,the effect of autophagy on cherry anthocyanin inhibition in cell lipid deposition was studied.It was found that cherry anthocyanins can further inhibit the deposition of HepG2 and L02 by promoting the formation of autophagosome,up-regulating the expressions of autophagy-related protein LC3,Atg5 and Beclinl,and down-regulating the expression of P62.Cherry anthocyanin can down-regulate the expression of fatty acid synthesis-associated protein FAS and ACC.This confirmed that autophagy was involved in the lipid-lowering effect of cherry anthocyanins.
