Adiantum flabellulatum L.is a folk herbal medicine.It is used internally in traditional Chinese medicine for the treatment of cold,fever,all kinds of inflammation,urinary system stones,external treatment of burns,snake bites and so on.It is reported that the leaves and rhizomes of A.flabellulatum contain multiple essential oils,and the methanol extracts of whole plant have analgesic effect.However,the effective components of A.flabellulatum have not yet fully clarified.In this thesis,the chemical composition and biological activity of A.flabellulatum were studied.The main results are as follows:1.Analysis of the content of flavonoids and phenolic acids in ethanol extracts of A.flabellulatum in two production areas of Guangdong and Guangxi.The plant materials extracted with 80%ethanol and determined the extraction rate.Using rutin and gallic acid as the standard,the content of flavonoids and phenolic acid in different parts of A.flabellulatum was determined by spectrophotometry.The DPPH,ABTS+free radical scavenging ability and reducing ability were determined in different parts of A.flabellulatum by spectrophotometry.The results showed that the maximum extraction rate was 16.4%in Guangxi.The contents of total flavonoids and phenolic acids in different parts of A.flabellulatum from different habitats were somewhat different.Aerial part of Guangxi had more contents,the total flavonoids content was25.53mg/g,and phenolic acid content was 39.25 mg/g.The DPPH and ABTS+free radical scavenging ability and reducing ability in the subterranean part of two places were higher.2.Analysis of the biological activity and chemical composition of different of A.flabellulatum.The plant materials extracted with pure n-hexane?HF?and ethyl acetate/dichloromethane?v/v=2:1??EF?of A.flabellulatum in Guangxi.The study of antioxidant activity showed that the scavenging ability of DPPH,ABTS+free radical and reducing abilities in the EF were higher than HF.The antibacterial activity study showed that the HF and EF had inhibitory effects on E.coli and S.aureus.The EF was more sensitive to S.aureus and MIC was less than 0.875mg/mL.The study on the activity of antirumor cells showed that high concentrations?360?g/mL?of HF and EF inhibited MCF-7?human breast cancer cells?.The chemical compositions of different extracts were analyzed by HPLC-MS.The results showed that the extracts of HF and EF contained a large number of secondary metabolite,including 52compounds such as flavonoids,phenolic acids,and fatty acids.3.Isolation and structural identification of chemical constituents of A.flabellulatum.Utilize silica gel column segmentation,MCI resin removal of pigment,MPFC separation,TLC separation and preliminary detection,recrystallization purification and other methods isolated from the HF and EF part of A.flabellulatum.The structures of 9 compounds were identified by 1H-NMR,13C-NMR,2D-spectrum,and mass spectrometry methods.The 6 of them were the first isolated from A.flabellulatum.,including D-?-T Cesium,locust-4-ene-6?-alcohol-3-one,?-sitosterol,4,4-dimethyl-1,7-pimelic acid,protocatechualdehyde,isoadiantol B.The other three are new compounds and they are flabellulatum 1,flabellulatum 2,flabellulatum3.Preliminary studies indicate that they belong to triterpenoids and fatty acid compounds,and their structures and activities are being identified and studied.This article has certain guiding significance to clarifying active components for A.flabellulatum,elucidating its medicinal value and further development and utilization.
