| "LiangTouJian" is the rhizome of Anemone raddeana Regel (Ranunculaceae), iscommonly used in Chinese folk medicine for curing rheumatism and neuralgia.In thisthesis, the investigation of chemical constituents and bioactivities of "LiangTouJian"arereported. The main research results are stated as below:1.After deely studying of the chemical constituents of "LiangTouJian", 24 compounds wereisolated.(All of them were elucidated on the basis of physico-chemical method, spectralanalysis and by comparison with some authentic samples. Among these compounds, two arenew compounds, They were elucidated as:raddeanosideR19(3-O-α-L-rhamnopyranosyl-(1→4)—-β-D-glucopyranosyl-(1→4)-α-L-arabinopranosyl-oleanolicacid28-O-β-D-glucopy-ranosyl-(1→6)-β-D-glucopyranoside(14);raddeanosideR20(3-O-α-L-rhamnopyanosyl-(1→2)-[β-D-gluco pyranosyl-(1→6)-O-β-D-gluco pyranosyl-(1→4)]-α-L-arabinopranosyl)-oleanolic acid 28-O-β-D-glucopy ranoside(15). And analyzed by GC-MS, The results of thechemical constituents of the oil compounds from column chromatography showed thatthirty-two compounds were isolated and identified.3..Bioactivities were deeply studied on the extraction and pure compounds from"LiangToujian".(1)Anti-tumor activitya.We preliminarily screened the total raddeanoside of "Liangtoujian" to S180 andHepAH.The results showed that the inhibitory rate of tumor of total raddeanoside wassignificant and but low.b.Comparison of the proliferous function to four kinds of cancers cell stub on refinedTRNS and pure compoundsThe results was that the extraction with three sugars or less(compound 11-13,sigle sugarchain with 3-aglycone) had more effective anti-tumor activity in vitro than those with moresugars(compound 14-16,double sugar chains with 3,28-aglycone)The inhibitory rate of tumor of refined TRNS was lower than that of TRNS. RefinedTRNS was attained after TRNS was decloured by D941 resin and enriched by D101 which made small polar compounds were lost. Further, the results showed that big polar compoundswith saponin of more sugers might go with lower or noneffective inhibitory rate of tumor in"Liangtoujian".(2) Anti-inflammation activity.We preliminarily screened the different extraction of Liangtoujian".The results were that theBu-OH and water extraction had anti-inflammation activity.4.Study of the pharmacognosy about"LiangToujian"Plant morphology, resource distribution and characters of "LiangToujian" werestudied. |
