Objective:Based on the establishment of asthmatic rat model,by interventing of total flavonoids of Adiantum capillus-veneris(TXJF,total flavonoid content>50%),to observe the general conditions of rats in each group and to detect the biochemical indexes and pathological changes of lung tissue,examine the effects of the TXJF on asthma model rats,then use the NMR spectroscopy(~1H-NMR)by measuring the NMR hydrogen spectrum to investigate the therapeutic effect of total flavonoids of Adiantum capillus-veneris(TXJF)on the asthma rats.Methods:Select the 60 healthy male SD rats,by OVA sensitization and atomization inhalation establish the model of asthma rats,randomly divided into normal group,positive control group(acetic acid dexamethasone group),asthma model group,total flavonoids of adiantum capillus-veneris(TXJF)group(high,medium,low dosage group)in each group has 10 rats,TXJF group were given to TXJF,then to observe the number and classification of inflammatory cells in broncho alveolar lavage fluid(BALF),the pathological changes of lung tissue,the determination of immunoglobulin E(IgE),interleukin-4(IL-4),and the concentration of interleukin-5(IL-5)in serum.Then observe the model rats weren't evaluated and the effects of TXJF on asthmatic models.Collect the serum and urine of each group rats and by NMR popper(~1H-NMR)detect the hydrogen spectra and establish the serum and urine metabolic fingerprint spectra of rats,and use to the orthogonal partial least square discrimination analysis(OPLS-DA)and the partial least squares discrimination analysis(PLS-DA)observe the difference of metabolic products in serum and urine of rats.Results:Compared with the normal control group,in asthma model groups rats were appear that asthma-like symptoms,the total number of cells and the percentage of inflammatory cells in the BALF were increased,the pathological changes in the lungs aggravated,and the level of serum IgE,IL-4,IL-5 concentration were increased.TXJF groups asthma-like symptoms were get better,the total number of cells and the percentage of inflammatory cells in BALF were reduced,and the pathological changes of lung tissue were reduced,and the level of serum IgE,IL-4,IL-5 in high and middle dose TXJF group was significantly lower than that in asthma model group.The results of serum ~1H-NMR detection PLS-DA analysis showed that the profile of the asthma model group and the normal group and the dexamethasone group and the TXJF intervention group were distinctly separated,compared with the normal group,the contents of leucine,glycine,creatine,glycoprotein,?-glucose and other compounds in asthma model group decreased significantly,and the content of lactic acid increased(P<0.05),the content of leucine,glycine,glycoprotein in the intervention group of dexamethasone acetate group and TXJF high,medium and low dosage groups were increased,the lactic acid content decreased(P<0.05),and the difference was statistically significant.Urine ~1H-NMR detection PLS-DA analysis results shows that the asthma model group was distinct from the normal and TXJF groups,and the TXJF group was close to the normal group.The contents of isoleucine,citric acid,succinate,glycine,alanine and other compounds in the urine of asthmatic model group were significantly decreased and the content of lactic acid increased.Compared with the model group,the content of TXJF groups,succinate,citric acid,creatine,taurine,alanine,glycine and so on in high dosage group increased,lactic acid and so on decreased obviously,the difference was statistically significant(P<0.05).Conclusion:TXJF has a certain therapeutic effect on asthmatic rats,it can improve the general situation of asthmatic rats,reduce the pathological changes of lung tissue,reduce the level of serum IgE,IL-4,IL-5,reduce the number and classification of inflammatory cells in bronchial alveolar lavage fluid(BALF),and has significantly effect on the serum and urine metabolites,and the mechanism may be related to the metabolic pathways of amino acid metabolism,energy metabolism and glucose metabolism in asthmatic rats.
