Objective:To clarify the absorption changes of crystal lanosine and deacetyl chlorosuccinic acid after different dosages of licorice processed by Morinda officinalis,in order to provide experimental basis for optimization of the processing technology of Morinda officinalis and to further research of the processing principle of making Morinda officinalis and lay the function.Methods:Morinda officinalis(the crude and 100:3,100:6,100:12,100:24 glycyrrhizic processed products)was extracted by water extraction and alcohol precipitation;HPLC method was used to determine the content of crystal lanosine and deacetyl chlorosuccinic acid;valgus intestinal sac method was used to explore the absorption characteristics of crystal lanosine and deacetyl acetylarginine;methanol was devoted to precipitate protein in these samples;the use of Originpro 8 software(Qt)was used to simulate the zero-order absorption equation,SPSS 21 was applied to calculate and analyze whether the absorption rate has a significant difference.Result:Absorption rate constants of different concentrations of crystal lanosine:The absorption rate constants of crystal lanosine in duodenum,jejunum,ileum,and colon were 1.40 ±0.32,1.43 ± 0.38,1.11 ± 0.35 and 1.11 ± 0.30;when the concentration of crystal lanosine in raw extract was 100.00 μg · mL-1;when the mass concentration was 200.00 μg · mL-1,the absorption rates constants of crystal lanosine in the duodenum,jejunum,ileum and colon were respectively 3.64 ± 0.76(P<0.01),35.85 ± 8.03(P<0.01),2.53 ± 0.86(P<0.05),and 25.16 ± 8.42(P<0.01);when the concentration was 400.00 μg · mL-1,the monotropein absorption rates constants in jejunum,ileum,and colon were 6.99 ± 1.15(P<0.01),6.73 ±0.78,3.43 ± 1.38(P<0.01),and 3.57 ± 1.29,respectively.Absorption rate ·constants of deacetyl acetylsuccinic acid with different mass concentrations:The absorption rate constants in the duodenumi,jejunum,ileum,and colon respectively were 0.69 ± 0.13,0.68 士 0.20,0.49 土 0.16,and 0.50 士 0.19;when deacetyl acetylsuccinic acid was found at a concentration of 51.07 μg · mL-1 in the extract.;when the concentration of deacetylographerin was 102.13 μg · mL-1,the absoption rate constants of the duodenum jejunum,ileum,and colon were 1.97 土 0.40(P<0.01),1.90 ± 0.42(P<0.01),1.27 士 0.40(P<0.01),and 1.20 ± 0.39(P<0.05);respectively;At 204.26 μg · mL-1,the absorptive rate constants of deacetylographic carolinate in the duodenum,jejunum,ileum and colon respectively were 3.07 ± 0.47(P<0.01),2.82 ± 0.69(P<0.01),1.90 ± 0.58(P<0.01),and 1.76±0.64(P<0.01).The absoiption rate constants of monotropein after different proportions of licorice processed:The absorption rate constants of monotropein in duodenum,jejunum,ileum,and colon in raw extracts were 3.64 ± 0.76,35.85 ± 8.03,2.53 ± 0.86,and 25.16 ± 8.42;100:3,respectively.The absorption rates of monotropein in duodenum,jejunum,ileum,and colon respectively were 4.42 ± 0.51,4.29 ± 0.49(P<0.01),2.73 ± 1.06,and 2.69 ± 0.97(P<0.01);100:6 product monotropein in the duodenum,jejunum,ileum,colon absoption rate Ka were 5.20±0.50(P<0.01),4.84±0.69(P<0.01),3.46±0.80,and 3.35±0.50(P<0.01);Absorption rate of crystalline lan blue glycosides in duodenum,jejunum,ileum,and colon were respectively 5.65 ± 0.91(P<0.01),5.61 ± 1.06(P<0.01),3.87 ± 1.31(P<0.01),and 3.54 ± 1.28(P<0.01);the absorption rate of crystal glucoside in the duodenum,jejunum,ileum,and colon of the 100:24 product was 4.97 ± 0.76(P<0.01),5.39 ± 0.95(P<0.01),3.48 ± 1.01,and 3.05 ± 1.14.The absorption rate constants of deacetylase carboglucuronide after different proportions of licorice processing result:The absorptive rate constants of deacetyl acetylarginine in duodenum,jejunum,ileum,and colon in raw product extracts respectivelywere 1.97±0.40,1.90±0.42,1.27±0.40,and 1.20±0.39;The rate constants of absorption of deacetylcarbenylsulfuric acid in the duodenum,jejunum,ileum and colon of the 100:3 licorice processing was 0.0250 ± 0.0018(P<0,05),respectively.2.50±0.18(P<0.05),2.55 ± 0.40(P<0.01),1,40 ± 0.54,1.44 ±0.60;Absorption rate constants of deacetyl carbenylsulfamate in the duodenum,jejunum,ileum,and colon of the 100:6 licorice processing respectively were 2.68 ±0.53(P<0.01),2.60 ± 0.48(P<0.01),1.49 ± 0.44,and 1.57± 0.57;Absorption rate constants of carboglucuronide in the duodenum,jejunum,ileum and colon of the 100:12 product were 3.11= 0.28(P<0.01),2.84 ± 0.32(P<0.01),2.01 ± 0.43,and 2.01 ± 0.35(P<0.01);Deacetylferrochelerylic Acid of 100:12 licorice processing in Duodenum,Jejunum,Ileum Absorption rate constants of colon and colon were 2.76 ± 0.33(P<0.01),2.24 ±0.51(P<0.01),1.83 ± 0.44,1.78 ± 0.41(P<0.05).Conclusions:In various intestinal sections were the linear absorption(R2>0.9),conformed to the zero order absorption rate.Monotropein and deacetyl acetylarginine in various intestinal sections were the linear absorption,conformed to the zero order absorption rate,indicating that both monotropein and deacetyl acetylarginine were passively absorbed.The higher the concentration,the faster the absorption rate of monotropein and deacetyl acetylarginine.The intestinal absorption rate constants of monotropein and deacetyl acetylarginine increased with increasing concentrations.With the increase of the proportion of licorice concoction,the absorption rate Ka of monotropein and deacetyl acetylarginine increased first and then decreased,reaching a maximum at the 100:12 licorice processing product.In addition to raw products,the absorption rate constants of monotropein and deacetyl acetylarginine in various bowel segments:duodenum≥jejunum>ileum ≥ colon.
