| Morinda officinalis How, which is one of the four famous southen medicines, is distributed widely in China and has been used in medicine, edible and health-care products. In order to promote the comprehensive utilization of Morinda officinalis How, the extraction and biological activites of major components from Morinda officinalis How have been studied in this thesis. The main experimental results are as follows:1. We detected the chemical components of Morinda officinalis How by pre-examination. The results indicate that Morinda officinalis How contains saccharides, anthraquinones, coumarin,amino acid, proteins, organic acid, flavones, volatile oil, steroids, hydroxybenzenes, probable contain cardiac glycosides, but no alkaloids, saponins. The contents of the important chemical constituents was also detected, there was 69.12% total saccharides, 5.84% polysaccharide, 0.28% polyphenols in the Morinda officinalis How.2. The Morinda officinalis How was extracted with different solvents, and the yield of extracts with water and ethanol was higher than that of using petroleum ether and ethyl acetate as solvent. The proper extraction conditions with water are as follows: the ratio of material to solvent 1:30, extraction time 1.5h, and temperature 60℃. The proper extraction conditions with ethanol are: the ratio of material to solvent 1:30, extraction time 1.5h, and temperature 60℃. Furthermore, the bioactivities of the extracts with different solvents were studied. The results showed that the scavenging rate of DPPH radical was 90.35% when the concentration of water extracts was 25mg/mL, and the scavenging rate of hydroxyl radical was 44.52% when the concentration of water extracts was 45mg/mL.3. The extraction of polysaccharide from Morinda officinalis How was further studied to determine the optimum extraction parameters. The optimum extraction conditions are as follows: water used as solvent, the ratio of material to water 1:30, extraction time 6h, and temperature 80℃. The yield of polysaccharide was 5.08% under the optimum conditions. The polysaccharide from Morinda officinalis How was purified, and its physical-chemical characteristics, chemical structure and constitunents were investigated. The results show that the polysaccharide from Morinda officinalis How is a polysaccharide with furan ring and composed of glucose, fructose and xylose. |
