ObjectiveTo investigate the effect of different concentrations of medicinal serum of Morinda officinalis polysaccharide on the expression of Integrin ?3,vacuolar H+ ATPase and tumor necrosis factor-? in cultured osteoclast system.MethodsThe mouse macrophage cell line RAW264.7 was induced with RANKL-inducing factor to form osteoclasts.The staining experiment of tartrate-resistant acid phosphatase and the bone sag formation experiment of bone-grinding tablets were carried out to identify the morphology and function of osteoclasts.Then the osteoclasts were divided into four groups.The control group,the medicinal serum of Morinda officinalis polysaccharide intervention group(treated with different concentrations of medicinal serum of Morinda officinalis polysaccharide for 48h).The mRNA and the protein expression levels of Integrin ?3,V-ATPase and TNF-? on osteoclast were respectively examined by real-time PCR and Western blot.SPSS 19.0 statistical software was used for one-way anova,and LSD was used for inter-group comparison.Results(1)Mice RAW264.7 cells were induced by RANKL for six days,then TRAP staining proved the formation of multinucleated osteoclasts and bone grinding tablets have bone resorption bursis.(2)The mRNA expression levels of Integrin ?3,V-ATPase and TNF-a on osteoclast in medicinal serum of Morinda officinalis polysaccharide treatment group were significantly lower than these in the control group(P<0.05).(3)The protein expression levels of Integrin ?3,V-ATPase and TNF-a on osteoclast in medicinal serum of Morinda officinalis polysaccharide treatment group were significantly lower than these in the control group(P<0.05).ConclusionThe medicinal serum of Morinda officinalis polysaccharide can reduce the expressions of Integrin ?3,V-ATPase and TNF-a on osteoclast.These suggested that the medicinal serum of Morinda officinalis polysaccharide can reduce osteoclastic bone absorption by decreasing the expression of Integrin ?3,V-ATPase and TNF-?.
