Iridoid glucosides,rich in Radix Morindae officinalis, have potent anti-inflammatory and analgesic activities,which is consistent to its traditional rheumatism function. In this experiment,main iridoid glucosides---monotropein and deacetylaspcrulosidic acid from Radix Morindae officinalis were isolatied and identified. HPLC chromatogram of monotropein and deacetylaspcrulosidic acid have been used to assign that of the original medicinal meterial.Simple and rapid methods was established to seperate and determine monotropein and deacetylaspcrulosidic acid from Radix Morindae officinalis,and enough reference substance was prepared for further experiments. The best adsorptive material on monotropein was find to purify monotropein, and influence Factors were studied when purifying monotropein by particle-reactive C. In addition, the stability of monotropein and deacetylaspcrulosidic acid were studied. The concrete content is as follows:(1) Seperate and identify main iridoid glucosides---monotropein and deacetylaspcrulosidic acid from Radix Morindae officinalis,and HPLC chromatogram of monotropein and deacetylaspcrulosidic acid have been used to assign that of the original medicinal meterial; Establish simple and rapid methods of preparation of monotropein and deacetylaspcrulosidic acid:Powder-reactive C was used to purify the main iridoid glucosides, then monotropein was crystalled from methanol solutions.A flash chromatography system on ODS was used for further purification,the result is that deacetylaspcrulosidic acid crystal precipitated at methanol-acetone solution of the purification.This study provided a foundation for more Research of the Quality Control on Morinda officinalis How and further Pharmacodynamic study.(2) Using monotropein as evaluation Indicators and HPLC as detection means,Adsorption of monotropein in Radix Morindae officinalis was studied by Different Macroporous Adsorptive Resins and active C. The results indicate that the particle-reactive C has good adsorption and desorption. Influence Factors were studied when purifying monotropein by particle-reactive C. And the best medicament concentration, adsorption temperature and elution solvent and so on,were studied. The result is 0.25g/ml ofmedicament concentration,80℃adsorption temperature,8h of the adsorption times,20% of elution solvent. The content of the monotropein in purified compound is more than 6%.(3) Effect of Purifications of Radix Morindae officinalis on the Cerebral Microvascular Endothelial Cell in Rats was studied. The level of BJT-1 can improve Energy of the BMEC obviously, BJT-2,BJT-3 and BJT-4 showed tendency of strengthening vitality of BMEC. Series of BJT-1,BJT-2,BJT-3 and BJT-4 can not resist injury of cerebral microvascular endothelial cells by hypoxia.(4) The stability of monotropein and deacetylaspcrulosidic acid were studied. The result is, the content of the monotropein and deacetylaspcrulosidic acid of Radix Morindae officinalis keep stable in a certain storage time; in neutral and alkalescence circumstance, the Structure of monotropein and deacetylaspcrulosidic acid were stable; when in acidic conditions,monotropein can converted to deacetylaspcrulosidic acid. The possible mechanism is that monotropein was dehydrated to become allylic ion,then rearranged and hydrated. Meanwhile,there were another hydrolysate determined.(5) Established determination method of the monotropein and deacetylaspcrulosidic acid, the method of HPLC was used with chromatographic conditions:Venusil MP C18 (4.6mm×250mm,5μm) column,the mobile phase was methanol-0.1% phosphate solution(5:95), the velocity of flow was 1ml.min-1,the detection wave length was 235nm,and the column temperature was 25℃.And determine the content of monotropein and deacetylaspcrulosidic acid of fifteen samples collected in Guangdong and Guangxi. The content of monotropein and deacetylaspcrulosidic acid in the crude Radix Morindae were 0.991%,0.432%;and there was greater difference on that of the produced.
