Objective:Uniform Design Experimentation was chosen to optimize the processing procedure of salted Morinda Officinalis.The parameters were consummated after the the optimal preparation conditions were confirmed.To compare the contents of oligose in different processed products with high-performance liquid chromatography,to discuss the efficacy of change in Morinda Officinalis and salted Morinda Officinalis according chemical constituent change.To make a comparison of Morinda Officinalis before and after salt processing procedure by pharmacology experiment.To research and evaluate the changes of brain function in Morinda Officinalis and salted Morinda Officinalis.Method:U6(63)uniform design table was used to investigate the four important factor in the salt processing procedure of salted Morinda Officinalis.Six level was designed for each single factor.These four factors are salt level in brine,infiltrating time,steaming time and Steaming power.Nystose was regarded as assessment index in the Morinda Officinalis after salt processing procedure.The chromatographic condictions as follow:Chromatographic column:Accusil C18(250mm×4.6mm,10?m);Mobile phase:Methanol-water(3:97);Column temperature:30?;Flow rate:0.8mLˇmin-1;Inject volumn:10?L;Tube temperature:80?;Atomization temperature:50?;Nitrogen pressure:35MPa;Gain:4.Content of oligose in different processed products were determined with the chromatographic condictions above.SPSS 19.0 statistical analysis software was applied to process independent t-test statistical analysis in Morinda Officinalis and salted Morinda Officinalis,and compare their difference.According the pharmacological experiment to contrast the difference of brain-strengthening effect in Morinda Officinalis and salted Morinda Officinalis.AlC13 was used to conduct brain nerve injury in rats to building Alzheimer's disease in mice model in short period.Water-extraction of Morinda Officinalis and salted Morinda Officinalis was given by gavage to evaluate the effectiveness of rats' brains were taken after 60d to Norepinephrine,dopamine,5-hydroxytryptamine in monoamine neurotransmitter were determined by cullen array potentiometric electrochemical detector.Brain-strengthening function of Morinda Officinalis before and after salt processing procedure by determining the content of three kinds of monoamine neurotransmitter.Results:1 The optimization of salted Morinda Officinalis preparation processAccording to the results of uniform design experiment,through stepwise linear regression analysis,The linear regression equation was obtained:Y=10.671+0.007X1-0.952X3-0.009X4,(P<0.05).According to the linear regression equation,the crucial factors of salt processing procedure of Morinda Officinalis were steaming time>salt level in brine>Steaming power>infiltrating time.Finally,the optimal preparation conditions were as follows:100 g Morinda Officinalis steamed 5.5 h under the heating power of 600 W after 8.5 h infiltrated by 120 mL salt water(with 2 g salt).Verification experiment results showed that the method is reliable by the RSD value was 0.67%.2 Comparison saccharides of Morinda Officinalis before and after salted processing procedureMonosaccharide composition of salted Morinda Officinalis was increased,while the content of sucrose,kestose,Nystose and f-fructofuranosylnystose was in different degree of decline.The paired t test was applied with ten batches of Morinda Officinalis and the corresponding batches of salted Morinda Officinalis.For a given level of significance 0.05,t(0.05/2,10)=2.228,The paired t test results showed that the score of fructose,sucrose,kestose,Nystose and f-fructofuranosylnystose were all<2.228,The analysis results were statistically significant(P<0.05).Significant changes have taken place in fructose,sucrose,kestose,Nystose and f-fructofuranosylnystose after salted processing procedure.3 The effects of Morinda Officinalis before and after salted processing procedure on the brainAn independent t test of the experimental data was proceed by SPSS 19.0.When contrasted with blank controlled group,the content of three kinds of monoamine neurotransmitter in rats' brain from model group were significantly reduce.It explained that rats' brain damage model was built successfully.Compared with model group,the content of three kinds of monoamine neurotransmitter in positive control group,high level group of Morinda Officinalis,and Low level group,medium level group and high level group of salted Morinda Officinalis.Independent t test was processed in Morinda Officinalis group and salted Morinda Officinalis group,The results showed that the content of three kinds of monoamine neurotransmitter in high level group of Morinda Officinalis and high level group of salted Morinda Officinalis had significant difference.Conclusion:1 The optimization of salted Morinda Officinalis preparation processUniform design method can be used for the optimizing of the processing procedure of salted Morinda Officinalis.Compare with the other statistical approach,Uniform design method can effectively reduce the number of experiments and improve the experimental efficiency.Uniform design method is More simple and convenient than the other methods,is able to effectively select the optimal preparation conditions.2 Comparison saccharides of Morinda Officinalis before and after salted processing procedureOptimal preparation conditions were as follows:100 g Morinda Officinalis steamed 5.5 h under the heating power of 600 W after 8.5 h infiltrated by 120 mL salt water(with 2 g salt).In the process of salt,Most of the sugars in Morinda Officinalis were decomposed to direction of the monosaccharide.By comparing sugar component changes,to further clarify the relationship between carbohydrate constituents and efficacy in Morinda Officinalis.The transformation of sugars is the reason why the medicinal properties transform to moderate and the tonifying kidney effect is enhance in salted Morinda Officinalis after salt processing procedure.3 The effects of Morinda Officinalis before and after salted processing procedure on the brainThis study according to preliminary compare with brain-strengthening function of Morinda Officinalis and salted Morinda Officinalis.The results discovered that the brain-strengthening function of both Morinda Officinalis and salted Morinda Officinalis can effectively improve.And the brain-strengthening function of salted Morinda Officinalis is stronger than Morinda Officinalis.
