The Establishment Of Neuralgia Model After Herpes Zoster In Rats And The Study Of Mitochondrial Ubiquitin Ligase MITOL In Postherprtic Neuralgia

?Objective?In this experiment,MITOL antibodies were given in the spinal scabbard of the 4th and 7th day after the establishment of herpes zoster in rats,and the role of MITOL in the PHN of rats was investigated by the test of their pain.?Methods?1.To establish the neuralgia model of SD rats after herpes zoster:to prepare the virus suspension with varicella zoster virus VZV infection of African green monkey kidney fibroblasts?cv-1?,and subcutaneous injection between the first and second webbed of rats.And a determination of the hot pain threshold?HWL?and the mechanical hyperalgesia threshold?PWT?of a SD rat was performed on day 0?D0?,1?D1?,3?D3?,4?D4?,7?D7?,14?D14?,21?D21?days.2.The role of MITOL at different time points in the PHN model of rats:the SD rats were randomly divided into4 groups?n=10?,and the model was formed.MITOL antibodies were injected into the experimental group on the 4th and 7th days respectively,and 0.9%normal saline was injected into the blank control group for the 4th and 7th days.The thermal pain threshold?HWL?and mechanical pain threshold?PWT?of D0,D1,D3,D4,D7,D14and D21 were measured.3.Statistical analysis:SPSS 21.0 software was used for statistical analysis,and the mean value addition and subtraction standard deviation?x?s?was used for the measurement data.The variance analysis of repeated measures was used at different time points,and t-test was used for comparison between groups,and P<0.05 was considered statistically significant.?Results?1.The PHN model of rats was established:there was no significant change in the thermal pain threshold and mechanical pain threshold of blank group B.Experimental group A disease 0 rats heat pain threshold and mechanical pain threshold are there is an obvious decline in day 3?P<0.05?,and in the seventh heaven to A minimum,recovered in 14 day,21 day more obvious but not made before the mold.2.MITOL at different time points of intervention in the role of PHN rats model:4 A disease,A disease in the two groups were 7 building success,on the third day its thermal pain threshold and mechanical pain threshold are significantly decreased?P<0.05??with A module?,to the lowest in the seventh heaven,recovered 14 days,and its thermal pain threshold in 21 days in the two groups have been restored to normal value?P>0.05?.The mechanical pain threshold did not return to normal.However,data analysis showed that the recovery of group A in the fourth day was better than that of group A ₇?P<0.05?.?Conclusion?1.The PHN model of rats was established successfully.The model is scientific and reproducible.2.MITOL plays an active role in PHN in rats.Early use of MITOL-antibody intervention can improve pain threshold in rats,relieve pain and recover early.
