| Background The incidence of dry eye menopause females is higher than young women and men,research think women menopause is one of the important influencing factors of dry eye disease.At present,a lot of simulation animal model of postmenopausal women in the majority with rat,rabbit,dog,establish to ovarian monkey animal model,dynamic observation of animal sex hormone levels in the body and the change of ocular surface related inspection,explore to ovarian monkey simulate dry eye,the feasibility of menopausal women.Objective Aim to elucidate the dynamic influence on level of animal sex hormone in the body,ocular surface osmotic pressure,Schirmer I test(S I t),corneal fluorescence staining and conjunctival impression cytology test after establishing ovariectomized monkey dry eye animal model.Methods Six healthy adult female cynomolgus monkeys were randomly divided into the ovariectomy group and control group evenly.Compared with monkeys ovariectomized,the control group had the same surgical procedure but bilateral ovaries removed.Venous blood of both groups from lower limb was gathered at preoperative and postoperative 1,3,6 month.Then,we investigated the concentration of serum estradiol and testosterone via ria method,ocular surface osmotic pressure,Schirmer I test(S I t),corneal fluorescence staining and conjunctival impression cytology test.Results Concentration of erum estradiol concentrations in ovariectomy group was lower than control group 3 months after operation.There was a significant difference(t = 7.64,p < 0.05).Whereas tears osmotic pressure tested at 3 and 6 months after operation was higher significantly(postoperative 3 m: t = 4.58,p < 0.05;postoperative 6 m: t = 6.82,p < 0.05).Moreover,there was no obvious difference in Schirmer I test of both groups at preoperative and postoperative 1,3,6 months.And,corneal fluorescent staining had not been accessed in two groups at any one point in time.In addition,we confirmed the conjunctival impression cytology examination in experimental group,the outcome showed that: The first month after operation: epithelial cells,0 level change,larger,multiform and easy to distinguish;goblet cells,number of cells increasing,PAS staining positive.The third month: epithelial cells,Ⅰ level change,larger and pleomorphic;goblet cells,number of cells decreasing,PAS staining positive.The sixth month: epithelial cells,Ⅱ level change,polymorphic,hard to distinguish;goblet cells,number of cells decreasing,blurry peripherally,PAS staining negative or weak.And in control group after operation,epithelial cells was 0 level change,and goblet cell number was increased,PAS staining positive.Conclusions We use cynomolgus monkey dry eye animal models to simulate dry eye menopausal women.It may be significant in researching the change tendency of the ocular surface and pathological mechanism of menopause women. |
