Pathological Image is an important basis for medical diagnosis,and treated as the gold standard for various diseases.However due to the reasons such as section making defection and the illumination differences,the generated pathological image is typically intensity inhomogeneous,edge blurry and lack of dynamic range,which directly affect the pathologist to make correct diagnosis.Aims at the problem above,we proposed a method to improve the quality of pathological image by extending the dynamic range and detail enhancement.The method can produce clearer pathological image for the pathologist,which has great research significance and practical value to the computer-aided diagnosis of pathological image.Based on studying and analyzing large amount of related domestic and foreign literature,with H&E(Hematoxylin and Eosin)staining pathological image as the research object,this paper mainly researched the high dynamic range(HDR)pathological image composition and HDR pathological image detail enhancement,and implemented a HDR pathological image enhancement system.(1)On HDR pathological image composition,firstly,we remove the intensity inhomogeneity of pathological image by color correction based on bias model and statistic information;then we simulate pathological image sequence with various exposure levels by different enhance method,and solve the camera response function by least square fit.Lastly we composite the high dynamic range pathological image.(2)On HDR pathological image detail enhancement,we take example by traditional detail enhancement based on bilateral filter,and proposed the HDR pathological image detail enhance method based on fast guided image filter.We use the fast guided image filter to separate the image into base layer and detail layer,and then compress the dynamic range of base layer with histogram mapping.And then take an additional step to suppress noise and enhance edge information by adaptive mask.Then the processed basic layer and detail layer are composited again to form the detail enhanced pathological image.In the end,we designed and implemented the HDR pathological image enhancement system based on module design pattern.The system implement the function of basic image enhancement,image sharpen,HDR pathological image composition and HDR pathological image detail enhancement.The system is full-featured and easy to operate.The experimental result about HDR Pathological image composition and detail enhancement shows that,the color correction algorithm we proposed in this thesis can remove the intensity inhomogeneity ideally.Meanwhile,the time validity and the effect of detail enhancement about HDR pathological image detail enhancement algorithm based on fast guided image filter have been preferably improved.
