Study On The Chemical Constituents And Antitumor Activitie Of Premna Fulva

Premna fulva is a species of Peremna linn family and a kind of deciduous shrub plant.It grows at the abdomen evergreen broad-leaved forest which altitude of 500 to 1200 meters with medicinal value such as anti-inflammatory effect,antioxidation and so on.Recently,terpenoids and flavonoids with excellent antibacterial action,antitumor and other activities were discovered by domestic and foreign scholars from this genus.In this topic,chemical constituents of P.fulva were studied and 27 compounds were obtained by the modern chromatographic techniques of chromatography on sillcagel,HPLC,Sephadex LH-20 and so on.Their structures were identified by physicochemical property,MS,CD and NMR spectra as ?-sitosterol(1),succinic acid(2),3?-acetyl-11?,12?-epoxy-olean-28,13?-olide(3),friedelin(4),friedelinol(5),ursolic acid(6),(+)-sesamin(7),piperonylic acid(8),oleanolic acid(9),naringenin(10),4-hydroxysesamin(11),apigenin(12),5a,8a-epidioxy-(22E,24R)-ergosta-6,22-dien-3?-ol(13),methyl-3,4-dihydroxybenzoate(14),asperglaucide(15),gummadiol(16),4,8-dihydroxysesamin(17),2?,3?-dihydroxy-urs-12-en-28-oic acid(18),sitoindoside I(19),9,10,13-trihydroxy-11-octadecenoic acid(20),machigline(21),vitexin(22),apigenin-8-C-?-D-xylopyranoside(23),cimicifugic acid(24),helichrysin A(25),(2R)-naringenin-5-O-?-D-xylopyranosyl(1?6)-?-D-glucopy-ranoside(26),(-)-syrin-gare-sinol-4,4'-bis-O-?-D-glucopyranoside(27).Compounds H-23 and H-26 are two new natural products.The antitumor activitites against to HL-60,Bcap37,SMMC-7721 and P388 of compounds H-11,H-15,H-17,H-23,H-26 and H-27 were examined by MTT assays.As a result,compound H-15 show better antitumor activity against to these four kinds of tumor cells with IC50 values of 21.36 ?g/mL,16.59 ? g/mL,35.24 ?g/mL,28.61 ?g/Ml,antitumor activity(IC50)of compound H-23 and H-26 against to HL-60,Bcap37 and SMMC-7721 were all less than 40 ?g/mL.This issue also summarized the chemical constituents and biological activities of Peremna linn.The results of this study will provide the scientific basis to the development and utilization of Premna fulva.
