Study On The Chemical Constituents And Antioxidant Activity Of Glochidion Puberum (Linn.)Hutch.

Plants of the genus Glochidion puberum(Linn.)Hutch.(Euphorbiaceae),widely distributed in south and southwest of China,are commonly used as folk medicines in the treatment of influenza,sore throat,dysentery,impaludism,rheumatoid arthritis,dyspepsia jaundice.The main constituents of Glochidion puberum have been revealed to be flavonoids,triterpenes,volatile oil,steroids,phenolic compounds,etc.Glochidion puberum L.Hutch exhibits a wide variety of biological and pharmacological activities,such as antimicrobial activity,anti-inflammatory and analgesic effects,antioxidant activity,antitumor activityGlochidion puberum was extracted by the method of reflow by ethanol.Then the extract was got by concentrating the filtrate.The extract was suspended by adding water to it and the suspension was abstracted by petroleum ether,dichloromethane,acetic ether,and n-butanol respectively,and each experiment was carried out for three times to get the ligarine part,the dichloromethane part,the acetic ether part,n-butanol part and the mother liquid of the water layer.These crude extract and compounds were assessed for DPPH free radical scavenging activity.Among these tested,the six screened parts exhibited significant radical scavenging activities on DPPH assay(IC50 value is25.24?g/mL?41.81 pg/mL?116.7?g/mL?8.62?g/mL?45.56?g/mL?44.66?g/mL g/ml).And they showed the stronger activity than the VC used with positive control(ICsovalue is 4.48 g/ml).According to the experiment,nine monomer compounds are finally obtained by separating ethanol extract of petroleum ether extract and ethyl acetate extract at the root parts of the Glochidion puberum through positive and reverse phase silica gel column chromatography and glucan gel column chromatography.The nine compounds:lupenone(1)and glochidone(2),3-epi-lupeol(3),glochidonol(4),3-epi-glochidiol(5),3a,23-dihydroxylup-20(29)-ene(6),glochidiol(7),catechin(8),(-)-gallocatechin(9)are analyzed and identified by modern spectroscopy methods such as nuclear magnetic resonance according to physical and chemical properties.The anti-oxidizing ability was tested by DPPH method and the IC50 value of the compounds were 3.43?M,3.78?M.According to the result the anti-oxidizing ability was listed as follows:acetic ether>petroleum ether>n-butanol>water layer>dichloromethane.9 compounds were dissected from the ethanol-aqueous extract of Glochidion puberum.
