Daylily flowers,the flower and bud parts of Hemerocallis fulva or H.citrina,are well known as Wang-You-Cao in Chinese,meaning "forgetone's sadness" plant.They have been recorded to produce antidepressant effects in ancient Chinese medicine.In this work,extraction and purification process of total flavonoids from daylily flowers were studied.The quality control method of daylily flowers were initially established.Based on screening and evaluation of the antidepressant active site,its working mechanism was discussed.1 Extraction and purification process of HF-total flavonoidsThe optimum extraction process is determined as:90%ethanol distillation for 2h.The extract of Hemerocallis fulva(HF)were separated with AB-8 resin into different factions containing 10%,30%and 50%ethanol.2 Quality control Study of HFUPLC method was established for rutin content determination in HF.Based on determination method,the UPLC method was established for HF-fingerprint.3 The mechanism of neuroprotective effect of HF-extractThe neuroprotective activity was determined using PC12 cell damage models induced by corticosterone,hydrogen peroxide and glutamate by evaluating the cell viability and lactate dehydrogenase release.The results showed that 10%,30%and 50%ethanol column chromatographic fractions of HF for the three models had better neuroprotective activity than the other components.The neuroprotective effect is possibly achieved by promoting cell proliferation and decreasing the release of LDH.4 Influence of HF-extract on neurotransmitters and its metabolites in nerve cellsThe LC-MS/MS detection was applied to investigate the change of DA,NE,ACh,GABA,Glu,TRP and their metabolite 5-HT,5-HTP and 5-HIAA in PC 12 cells.Experiment results showed that HF-10%,30%,50%ethanol column chromatographic fractions can improve dysfunction of neurotransmitter in model cells,and also reverse changes of neurotransmitter content.In conclusion,this study found that HF-extract has shown protective effect in depression models and this mechanism might be associated to maintenance of neurotransmitter homeostasis.This work laid a foundation for further studies to investigate antidepressant effect and provided a scientific basis for development of new antidepressant prescriptions based on HF-flavonoids.
